Abstract


Objective:To research on expression and function of miRNA-132in colonic implanted carcinoma in nude mouse.Methods:①A BALB/c nude mouse model of colonic implanted carcinoma was established by using tumor cell HCT-116and was divided into three groups randomly blank control group;DMSO group(dimethyl sulfoxide,DMSO) and inhibitor group(WP1066, the inhibitor of STAT3+DMSO).②The tumors will be harvested, then tested by real-time PCR for relative quantitative study for miRNA-132;③The mRNA transcription level of MMP-2,MMP-9was determined by real-time quantitative PCR and the expression of MMP-2,MMP-9were tested by immunohistochemistry staining.Results:Tumor sizes of blank control group and DMSO group had no significant difference (p>0.05). However the size of inhibitor group was significantly smaller than other two groups (p<0.05).The expression of miRNA-132has significant difference between blank control group and inhibitor group (p<0.05).The mRNA transcription levels of MMP-2,MMP-9in tumor tissues had no significant difference between blank and DMSO group (p>0.05).However, there were significantly lower level in inhibitor group than other two groups (p<0.05).The protein expression of MMP-2,MMP-9had no significant difference between blank and DMSO group. However, there were significantly lower level in inhibitor group than other two groups(p<0.05).Positive correlation of mRNA transcription and protein expression level of MMP-2with MMP-9were detected respectively in implanted tumors of nude mouse, so did transcription of miRNA-132with mRNA transcription of MMP-9(P<0.05).Conclusion:miRNA-132could regulate positively growth of colonic implanted carcinoma in nude mouse established using tumor cell HCT-116. whileWP1066, the inhibitor of STAT3, suppress lymphatic generation, downregulate transcription of miRNA-132and mRNA transcription of MMP-9, so slower the growth rate of implanted tumor in nude mouse.
