Abstract


Background and objectivesWith the improvement of people’s living standards, colorectal cancer (CRC) is not only one of the most common gastrointestinal malignant tumors in the world, but also obesity-related tumors. In China, form and development of colorectal cancer is causes of high-protein, low-fiber diet, and less exercise. the prevalence of colorectal carcinoma has increased year by year in the past several years. Until now, the colorectal cancer mortality rate has been ranked the fifth, and incidence rates has been ranked the fourth. Beijing and Shanghai is even higher. The complex process of tumor development is a multi-gene changes and multi-step carcinogenic. It is generally believed that the normal cell development from hyperplasia to malignancy is a gradual evolution:general hyperplasia→dysplasia→cancer. Colorectal cancer is most from pre-existing adenomas. This process requires a longer time, so the early blocking colorectal developed as an effective method of treatment. The various approaches of clinical treatment is including surgery, chemotherapy, radiotherapy, neoadjuvant chemotherapy and molecular targeted therapy. Due to the fact that most colorectal cancer patients are in the terminal stages of cancer when they receive treatments. Hence, it is the key to treat the malignant tumors by detecting early, diagnosising early, treating early.Adiponectin is an endogenous bioactive factors secreted by fat cells specific widely distributed in a variety of human tissue cells. Closely related to coronary heart disease, diabetes, high blood pressure, cancer and other diseases. It is capable of regulating glucose and lipid metabolism, improve insulin resistance, antiinflammatory, antitherosclerotic and anti-tumor effects. Adiponectin plays a role in by adiponectin receptor binding, and its receptors are divided into two structures:AdipoRl and AdipoR2. Due to the sensitivity of adiponectin, so the different adiponectin and adiponectin receptor binding is to play the corresponding function. This combination is not only related to sugar and fat metabolism process, and are closely linked with the onset and progression of colorectal. Recently, a few study have showed that Rland R2expresses widely in various kind of cancer cells including colon cancer, prostate cancer, ovarian cancer, and hepatoma. But no experiments confirmed the expression of R1and R2in human colon carcinoma and colorectal adenomas comparison. In this study, we preliminarily confirmed that R1and R2was expressed in human colorectal cancer tissues and colorectal adenoma tissues and investigated the relationships between the expression of R1and R2and clinicopathological parameters of colorectal cancer. In order to treat the malignant tumors by detecting early, diagnosising early, treating early.Materials and Methods1Materials:We collected specimens from the First Affiliated Hospital of Zhengzhou University between April2012and July2012by surgical resection including51cases of colorectal cancer specimens,42cases of colorectal adenoma specimens and35cases of normal mucosa tissue.51cases of colorectal cancer specimens is consist of30men and21women who aged24to82(average47.8±11.9years). And the colorectal cancer patients were composed of15tubular adenocarcinoma,13papillary adenocarcinoma,13mucinous adenocarcinoma and10signet-ring cell adenocarcinoma. With regard to the differentiation grades,10were graded as high, 13were graded as middle and26as low. According to staging system referring to UICC TNM Classification criterion, version sixth (2012),9patients were staged as I,24patients were staged as Ⅱ,16patients were staged as Ⅲ and2patients were staged as IV. There are14cases of lymph node metastasis and37cases without lymph node metastasis. In addition, the colorectal adenoma patients were composed of19tubular adenomas,16villous adenoma,17tubulovillous adenoma. All of the specimens were fixed in10%formaldehyde, paraffin embedded,4μm serial section for SP immunohistochemistry.2Main reagent:polyclonal rabbit antihuman Adiponectin antibody, S-P agent box, DAB color kit.3Methods:Immunohistochemistry Adiponectin was performed on cases using R1antibody at1:300ratio, R2antibody at1:250ratioand, PBS was used as a negative control.4Statistical analysis All statistical analyses were performed using the statistical package SPSS version17.0. Adiponectin acceptors expression levels of colorectal cancer were compared with those of colorectal adenoma and normal mucosa using Chi-square test. Applications listed contact number correlation analysis the expression of R1and R2with the three groups of colorectal tissue patients. Results were considerable statistically significant at the level of a=0.05.Results1The R1expression in three groups of colorectal tissue:Adiponectin acceptors was wildly expressed in all colorectal carcinoma tissues, colorectal adenoma tissues and normal colorectal tissues and mainly located in cytoplasm, which was pale-yellow, tan-yellow, brown grain. The strong positive rate and the positive rate of the expression of R1was49.0%(25/51) and19.6%(10/51) in colorectal carcinoma tissues. The weakly positive rate and negative rate of the expression of Rl was all15.7%(8/51) in colorectal carcinoma tissues. The strong positive rate and the positive rate of the expression of R1was21.4%(9/42) and35.7%(15/42) in colorectal adenoma tissues. The weakly positive rate and negative rate of the expression of Rl was19.0%(8/42) and23.8%(10/42) in colorectal adenoma tissues. The strong positive rate and the positive rate of the expression of Rl was14.3%(5/35) and17.1%(6/35) in normal colorectal tissues. The weakly positive rate and negative rate of the expression of Rl was48.6%(17/35) and20.0%(7/35) in normal colorectal tissues. The three groups is statistically significant.(P=0.001)2The R2expression in three groups of colorectal tissue:The strong positive rate and the positive rate of the expression of R2was45.1%(23/51) and21.6%(11/51) in colorectal carcinoma tissues. The weakly positive rate and negative rate of the expression of R2was19.6%(10/51) and13.7%(7/51) in colorectal carcinoma tissues. The strong positive rate and the positive rate of the expression of R2was16.7%(7/42)and40.5%(17/42) in colorectal adenoma tissues. The weakly positive rate and negative rate of the expression of R1was21.4%(9/42) and21.4%(9/42) in colorectal adenoma tissues. The strong positive rate and the positive rate of the expression of R2was17.1%(6/35) and20.0%(7/35) in normal colorectal tissues. The weakly positive rate and negative rate of the expression of R1was40.0%(14/35) and22.9%(8/35) in normal colorectal tissues. The three groups is statistically significant.(P=0.000)3The relationships between the expression of Adiponectin and clinicopathological features were examined. The expression levels of R1and R2in colorectal carcinoma tissues were correlated with tumor differentiation degree, lymph node metastasis and TNM stage (all P<0.05), but no significance relation with gender, age, or tumor size (all P>0.05).ConclusionsAdiponectin receptors R1and R2existed in the three groups of tissues. Furthermore in colorectal cancer and adenomas tissues the expression of adiponectin receptors R1and R2was enhanced. Adiponectin receptors expression is closely related with the degree of malignancy cancer.
