Abstract


An integrated methodology from micro-scale strains screening to large-scalebiomass production of microalgal bioenergy was built up in this study. A96-well microplates swivel system (M96SS) was designed for microalgalstrains screening for bioenergy utilization. The concordance of M96SS onstrains screening versus traditional standard flasks culture screening wasverified. Critical problems of intra-well mixing and evaporation loss wereimproved by sealed vertical mixing of the M96SS. The reliability andefficiency of the M96SS were verified through a comparison of traditionalflask culture and previous microalgal miniature culture methods. During thescreening of6microalgal strains, Chlorella sp. SJTU-3was selected asresearch material for further large-scale biomass production because of itshigh biomass yield in artificial medium (1.229g L-1) and also goodperformance in wastewater medium under20%CO2. Illumination wasdetermined as the main limitation factor for the raise of biomass yield ofChlorella sp. SJTU-3in its small-scale cultivation (500mL) in air-lift columnphotobioreactors. Finally, based on the data of preculture,9000lux double-side illumination and10%CO2concentration were selected asoperation conditions for culture optimization in pilot-scale (160L) tubularphotobioreactor. The modified biomass yield (7days culture) was1.05g L-1,which was2.5times higher than that in preculture.
