Abstract


The ticks are obligate blood-sucking ectoparasites widely distributed, its spread of human and animal diseases are very great harm to humans and livestock development. Ticks highly reproductive, of vitellogenesis its hormonal regulation is a hot research direction in the field, provides a solid theoretical foundation for the biological control of ticks. Haemaphysalis longicornis longicornis study to study the morphology and protein content of ovarian development process, and the use of the vitellin monoclonal antibodies prepared in this laboratory, using ELISA technology, research and 20E (20-hydroxyecdysone ) and JH III (juvenile hormone III) the role of hormones on the the Haemaphysalis longicornis Vitellogenesis process of regulation. The main results are as follows: the stereomicroscope, ovarian development was observed characteristics, shows that with the bloodsucking, ovarian consequent significant changes. Mating season, the ovary is very fine, white. The ovarian blood meal thicker longer. 3 to 4 days after the blood meal, egg fast development, enlarged ovaries, oocytes brownish red, yolk granules appear. Five days after the blood meal to lay eggs, ovarian appearance of blood meal, four days after similar, but can be seen in the ovarian cavity and fallopian tubes are discharged egg. Eight days after spawning, ovaries begin to shrink, thinning white, only few are not discharged egg. The same time, the ovarian weight change significantly the ovary of the mating season is very light (0.867mg) and maximum weight 30.540mg to spawning the day of ovarian then begin to decline, down to a minimum 3.667mg to eight days after spawning ovarian weight. Accordingly, to the spawning two days of ovarian weight accounted weight ratio reaches a maximum 0.225 percent, after slowly decline, eight days after spawning dropped to 0.041%. The research results show that the injected dose 10μg/tick 20E and ticks 10μg/tick JH III died four days after injection. Injection dose 2μg/tick 20E ticks ovary was filled with egg yolk granules filled ovarian volume, as a whole showed brownish yellow, and the injection control ovarian tiny, white translucent. The 20E injection dose 2μg/tick ticks ovarian weight to body weight ratio, fat body, haemolymph and ovarian yolk white original protein content and vitellogenin protein content of the control group compared to significantly higher. JH III unmated half full blood longhorn the Haemaphysalis ovarian weight, fat body, haemolymph and ovarian yolk content and no effect on the content of vitellogenin. Applied on the day of the blood meal, the female ticks injected 20E and local JH III, 4 days weighing the ovarian weight ELISA assay fat body, hemolymph vitellogenin and yolk protein content in ovarian tissue, applied to the results for the hormone group and control group compared to no significant difference. Therefore concluded, 20E significantly promote ovarian development unmated female ticks half full of blood, stimulate vitellogenesis, but no effect occurred JH III unmated female ticks yolk half full of blood. 20E and JH III longicornis yolk blood meal period had no impact.
