Abstract


MCM-41mesoporous molecular sieve is often used as the carrier of drug delivery due to its large specific surface area and pore volume, one-dimensional uniform channel, hexagonal ordered arrangement, stable framework structure, easy modification, non-toxic, and so on. It opens up a new direction for mesoporous material in the application of drug delivery. Insulin as a kind of protein peptides hormone drug, it has short half-life, easy degradation, low bioavailability. Therefore, it is important to research sustained release preparation of insulin.In this paper, different MCM-41mesoporous molecular sieves were synthesized by hydro thermal method and room temperature method. MCM-41which was synthesized by hydrothermal method was modified by trimethylchlorosilane and3-aminopropyl-triethoxysilane. Porcine insulin as model drug was doped inside the pore channels of different molecular sieves via liquid-phase grafting method. The structures of the prepared materials were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and N2adsorption-desorption. And the author studied the release process of prepared drugs in a simulated body fluid solution. The release results showed that amination MCM-41as carrier sample (AM/Ins) showed the best release effect. AM/Ins showed a sharp release burst in initial1h. At12h, the cumulative release rate of AM/Ins reached48.07%. The cumulative release rate of AM/Ins only reached79.42%at32h, and it reached99.09%at38h.
