Paeonia otsii, the wild relative of cultivated peony, belongs to the deciduous sub-shrub of Paeonia. The seed oil of P. ostii contains abundant unsaturated fatty acidswith a very important healthy and medicinal values. In March2011, the Ministry ofHealth authorized the seed oil of P. ostii and P. rockii as a new source of applied food(Ministry of Health of the People’s Republic of China in2011, the9thannouncement),and the seed oil of P. ostii has been for sales on the market currently. P. otsii cangenerate a lot of seed cake through oil extraction, which has attracted great researchand development interests. Mang seeds species of Paeonia’s chemical constituents andpharmacological activities have been studied. However, to the best of our knowledge,the research on chemical constituents and pharmacological activities of seed cake fromP. otsii remains an ongoing challenge.Our previous studies have shown that the seed cake of Paeonia otsii has anantibacterial effect, in which n-butanol fractionation has strong activities. The researchemploys preparative HPLC, Sephadex LH-20and column chromatographic methods toprepare isolation and purification, and then identifies the compound structures usingspectral data analysis. Fifteen compounds were isolated from the seed cake of Paeoniaostii, including8monoterpene glucosides,4stilbenes and3other compounds. Thesecompounds are:(1) albiflorin R1,(2) paeoniflorin,(3) oxypaeoniflorin,(4) albiflorin,(5)4"-hydroxyl-albiflorin,(6)1-O-β-D-glucopyransoyl-paeonisuffron,(7)6’-O-β-D-glucopyranosylalbiflorin,(8) β-gentiobiosylpaeoniflorin,(9) suffruticosol A,(10)suffruticosol B,(11) suffruticosol C,(12) polydatin,(13)7-O-β-D luteolin glucoside,(14) sucrose,(15)1-(p-hydroxybenzoate)--D-glucopyranose(1→6)--D-glucopyran-osyl. All of compounds were isolated from the seed cake of P. ostii for the first timeand compound15were identified as a new compound.Antibacterial activity experiment showed that n-butanol extraction fromthe seed cake of P. ostii has obvious antimicrobial action. Stilbene compounds9-11 have strong inhibition activity. This research provides a scientific basis for looking fornew antibacterial substances.
