Abstract


Objective: The main topic is divided into four parts, from in vivo and in vitro studies of vitamin D infection immune regulation. Mouse monocyte macrophage cell line-RAW264.7, to study 1,25 - dihydroxyvitamin D (1,25 (OH) 2D3) on the function of macrophages after LPS stimulation and of itself apoptosis; Second, the use of mouse monocyte macrophage cell line-RAW264.7 study of 1,25 (OH) 2D3 on Staphylococcus aureus phagocytic activity of macrophages, and reactive oxygen species generation, and their own apoptosis; three using C57/BL mice, 1,25 (OH) 2D3 on LPS-induced acute lung injury, the release of inflammatory cytokines mice, the mice survival time after the occurrence of pulmonary edema and septic shock in mice; IV method of case-control study of the relationship between Research Machine vitamin D metabolite levels and diabetes in patients with lung infection. Method: First, macrophages supplement of 1,25 (OH) 2D3 24h, detected by flow cytometry of bacterial lipopolysaccharide LPS-induced macrophage reactive oxygen species generation, the rate of apoptosis, mitochondrial transmembrane potential and intracellular free calcium change of the ion concentration; macrophages supplement of 1,25 (OH) 2D3 24h, using flow cytometry Staphylococcus aureus (Staphyloccocus aureus Rosenbach SA) infected macrophages to swallow the bacteria activity, the generation of reactive oxygen species , mitochondrial transmembrane potential and intracellular free calcium concentration changes; three, C57/BL mice subcutaneous injection of 1,25 (OH) 2D3 after a week, to different doses of LPS infection to establish acute lung injury and septic shock animals model, ELISA assay mice serum, cardiac and lung homogenates content of TNF-α, lung wet / dry weight (W / D) detected pulmonary edema, and occurrence of the survival of mice with septic shock time survival analysis; diabetes complicated by lung infection for the study groups, case-control study the relationship between serum vitamin D metabolite levels and lung infections. Results: LPS-induced experimental results show that the supplement of 1,25 (OH) 2D3 group macrophage apoptosis rate was significantly lower (P lt; 0.05); mitochondrial membrane potential in 1h, 3h significantly high (P lt; 0.01 ); intracellular calcium ion concentration is low but not statistically significant difference; release of active oxygen in 3h was significantly higher (P lt; 0.05), 1h, 6h significantly lower (P lt; 0.01), and the membrane potential and withered mortality, active oxygen exists a good correlation between the concentration of calcium ions. Staphylococcus infections experimental results show that supplemental 1,25 (OH) 2D3 phagocytic activity of macrophages 1h significantly enhanced (P lt; 0.01), 6h significantly lower (P lt; 0.01); release of reactive oxygen species 1h significant higher (P lt; 0.01), 3h, 6h significantly with lower (P lt; 0.01); mitochondrial membrane potential 1h significantly lower (P lt; 0.05), 3h significantly with increased (P lt; 0.05); intracellular calcium concentration 1h significantly lower (P lt; 0.01). And phagocytic activity and reactive oxygen species, respectively, the concentration of calcium ions show a good correlation. Septic shock experimental results show that half of the time of death of the supplement 1,25 (OH) 2D3 and pure LPS model group respectively for 65h and 100.5h,. Acute lung injury in experimental lung W / D intravenous injection of LPS2.5mg/kg, to 6h supplement of 1,25 (OH) 2D3 lung W / significantly lower than LPS group (P lt; 0.05); Serum The concentration of TNF-a injection of LPS 5.0mg/kg 2h after the VD LPS group than LPS group was significantly higher (P lt; 0.01) LPS 2.5mg/kg 2hVD, LPS group was significantly lower than LPS group (P lt; 0.01) , 6h was significantly higher than that in LPS group (P lt; 0.01); injection of LPS 5.0mg/kg TNF-a concentrations in the lung tissue after 2 hours the VD LPS group than LPS group was significantly higher (P lt; 0.05), LPS 2.5mg / kg, 2hVD LPS group was significantly higher than the LPS group (P lt; 0.01) lower than LPS at 6h group (P lt; 0.05); myocardial tissue concentration of TNF-a after LPS injection supplement 1,25 (OH) 2D3 group compared with LPS group, there are different degrees of elevated 5.0mg/kg, 2h and 2.5mg/kg, 6h was significantly higher than that in LPS group (P lt; 0.05, P lt; 0.01). Diabetes patients with concurrent infection GHA1C (%) gt; 10, serum 25 (OH) D ≤ 20 ng / ml, serum 1,25 (OH) 2D3 ≤ 20 pg / ml relevant. Conclusion: 1,25 (OH) 2D3 to enhance macrophage defensive role in the outer-borne infections. 1,25 (OH) 2D3 to extend the survival time of mice with immunodeficiency, relieve pulmonary edema. Three patients with diabetes mellitus, low serum levels of vitamin D is a risk factor for pulmonary infection.
