Phellinus igniarius is a precious medicinal fungus, its secondary metabolites flavoneshas high medicinal value. The means of multi-scale was used to enhance the production oftotal flavones. The fermentation parameters were optimized by Response SurfaceMethodology. Based on the optimized fermentation conditions, a new fermentation processtwo-phase culture was established. Finally effect of three precursors and two abiotic elicitorson the production of total flavones was studied.The fermentation parameters were optimized to increase the total flavones productionusing Plackett-Burman design and Response Surface Methodology. The experimental resultsindicated that the optimized composition of medium was20.00g/L Glucose,20.00g/LMaltose,20.00g/L Mannitol,9.70g/L Corn Powder,8.74g/L Yeast extract,5.00g/L NaCl,0.50g/L FeSO4,0.05g/L CuSO4,1.50g/L VB1, and the total flavones was1532.83μg/ml.The optimized experimental conditions were: pH6.0; temperature30°C;100ml mediumin250ml shake-flask; rotation speed150rpm; inoculation volume10%; seed age8-9d.Two-phase culture, a new fermentation process was established using a size of macro porousresin. During the process, flavones was first selectively adsorbed to the resin then eluted byeluent, and the total flavones was2174.35μg/ml.Effect of three precursors (phenylalanine, cinnamic acid and sodium acetate) and twoabiotic elicitors (silver nitrate and glutathione) in fermentation for flavones was studied. Theresults showed that both precursors and abiotic elicitors could promote the flavonesbiosynthesis. The positive effect was dependent on their concentration and addition time.When the two abiotic elicitors were combined to added, the maximum flavones productionwas1.42times as that of the control, more productive than they were added respectively.
