Abstract


Normal inhabited environment of marine microorganism is water body form marine.Comparing the conditions with dry land, high saline salinity, high pressure of water body, lowtemperature and fewer nutritions is the feature of marine. Because of these features of livingenvironment, comparing with terrestrial microorganism, marine microorganism hastremendous differences. Including physiological properties and physicochemical properties.Marine microorganism has special physiological properties, therefore marine microorganismcan produce new construction and muti-function active material. How to exploit the resourceis the focus of modern world. How to utilize the new active material is the topic of people.The theme of this thesis is to find the optimal components of culture medium and theoptimal fermentation conditions of culture bacteria strain. The bacteria strain D-11isdegrading bacteria of algin, the bacteria D-11is isolated form rotten kelp. The sample wascollected from the kelp farm of Liangshuiwan of Dalian Golden Pebble Beach. In this thesis,according to the DNS method, the optimal fermentation conditions and the optimalcomponents of culture medium of marine bacteria strain D-11was definite. The optimalcomponents of culture medium are sodium alginate0.4%, corn steep liquor1.85%, yeastextract paste0.92%, FeSO40.01%, sucrose3%, NaCl3%. The optimal fermentationconditions culture temperature28℃,the optimal pH6.0, the culture time4d, inoculumconcentration9%, content of culture medium volume30ml. Utilized the Response Surfacemethod analyzing the result of significant factors. The significant factors are corn steep liquor1.85%and yeast extract paste0.92%. Following the optimal components of culture mediumand the optimal fermentation conditions. The maximal enzyme activity of bacteria strain D-11is57.65U/ml.
