Abstract


Sedum lineaare Thunb is perennial herbaceous plant widely used as traditional botanical medicines. In recent years, pharmacological research displays that Sedum lineaare Thunb has great effects of anti-fatigue, against liver injuries, anti-lipid peroxidation, against cerebral and myocardial anoxia as well as anti-inflammatory. Especially, the total flavonoids of Sedum lineaare Thunb showed a remarkable antitumor effect. Chemical constituents of traditional Chinese medicine is the material basis of efficacy. In order to further study its biological activity, structure-activity relationships, metabolic, structure reformation and artificial synthesis, chemical structure of constituents which are got by the process of extraction, separation and purification, must be clear. This paper focuses on the chemical constituents of petroleum ether extraction part of Sedum lineare Thunb, totally8compounds were isolated by the various chromatographies over silica gel column, preparative thin layer chromatography, Sephadex LH-20, and identified by physical and chemical methods, EI-MS, IR,1H-NMR,13C-NMR spectral technologies. At present, the main methods of extraction and purification of total flavonoids from Sedum lineare Thunb are alcohol heating refluxing and macroporous resin adsorption by D101, which has several disadvantages in long extraction time, the large amount of solvent and low adsorption capacity of macroporous resin. The ultrasonic extraction of the total flavonoids from Sedum lineaare Thunb and the separation and purification process of the total flavonoids is not yet reported. The method of orthogonal design was first used to investigate the ultrasonic extraction condition. The purification effect of the total flavonoids by different macroporous resins was also studied together with experimental studies on scavenging activity of total flavonoids to DPPH free radicals.1. The chemical constituents analysis:Totally8compounds were isolated by the various chromatographies over silica gel column, preparative thin layer chromatography, Sephadex LH-20, and identified by physical and chemical methods, and modern spectral technology. They were①Tritriacontane,②δ-Amyrone,③δ-amyrin,③β-Sitosterol,⑤7-oxositosterol,⑥7a-hydroxysitosterol,⑦Daucosterol palmitate,⑧Daucosterol.2. Extraction of total flavonoids:Optimum ultrasonic extraction conditions (ethanol concentration, solid/liquid, ultrasonic time, working temperature) were obtained through orthogonal design. The optimum ultrasonic extraction conditions were80%ethanol, solid/liquid1:40, ultrasonic20min at30℃. The extraction rate of total flavonoids reached1.66%.3. Purification of the total flavones:The adsorption ratio and eluting ratio of total flavones were worked as an index. Through the static adsorption experiment, AB-8resin showed good absorption and purification capability of the total flavonoids from Sedum lineaare Thunb. The adsorbed AB-8macroporous resin column optimum eluting condition was washed by80%ethanol at the eluting velocity of2mL·min-1, the eluting ratio and purity of total flavones was94.72%and43.51%.4. DPPH free radical scavenging efficiency of flavonoids from Sedum lineaare Thunb:Contrasting to the VitC and the total flavonoids, study on DPPH scavenging capacity of total flavonoids from Sedum lineaare Thunb. Within tested concentrations, the results showed that the density of the total flavones and VitC were both presented dose-effect relationship with the eliminating ability of free radical. At the concentration of40μg·mL-1, the DPPH· clearance rate was82.31%which was more significant than Vit C.
