Abstract


Objective to study the of different concentration of EGCG solution onbladder epithelial cells of normal mice,to explore suitable experimental drugconcentrations and validate feasibility of mouse bladder instillation,Methods36Healthyfemale mice were randomly divided into7groups, the PBS group and,0.5mmol/L EGCG,1mmol/L EGCG,2mmol/L EGCG,4mmol/L EGCG,8mmol/L EGCG groups, each groupcontained6mice.perfused PBS buffer, and different concentrations of EGCG sterilesolution into the bladders of mice of corresponding groups,After24h, obtained specimens,performed pathology HE staining and fluorescence staining and detected their level ofexpression of inflammatory factors. Results Pathological HE and fluorescent stainingshowed that urothelium was observed with no apparent injury in PBS group and allEGCG group except8mmol/L EGCG group.,with no abnormal change,expression ofinflammatory factors in each group were no statistically significant differences inlevels.conclusions EGCG solution at a concentration that is less than or equal to the0.8mol,will have no significant impact on function and morphology of bladder urothelialcells, aund it can be considered a suitable intervention. Objective: to study preventive protection effect of EGCG against bladderinfection induced by Escherichia coli,And to assess its effect of resistance against Escherichia coli. Methods:According to bladder instillation methods, perfused differentconcentration of EGCG solution into the bladder in healthy mice, established4groups:0.5mmol/L EGCG,1mmol/L EGCG,2mmol/L EGCG,4mmol/L EGCGrespectively.and established PBS group. Then Perfused different concentration of EGCGsolution into bladers of corresponding Group of mice.After24h,50μl highly pathogenicEscherichia coli with the concertration of1x108cfu/ml was slowly perfused into thebladder transurethrally. At24hours post-perfusion of escherichia coli, HE staining andimmunofluorescence staining of bladder tissue were performed to observe the injury ofbladder epithelium and the expression level of inflammatory factor Interleukin-6(IL-6) andgranulocyte colony-stimulating factor (G-CSF) as well as assess the prevention effect ofEGCG in Escherichia coli induced acute bacterial infection in mice.Results: the degree ofinjury and the expression level of inflammatory factors were decreased in the PBS,1mmol/EGCG,2mmol/L EGCG,4mmol/L EGCG groups consecutively, and the4mmol/LEGCG group had the best prevention effect (P<0.05) injury was observed obviously in the0.5mmol/L, its expression of inflammatory factors compared with the PBS group with nostatistically significant differences.Conclusions: with a good antibacterial effects, EGCGhave a protective effect on urinary bladder epithelium. Methods:30healthy female mice were randomly divided into5groups:PBS group,1mmol/L EGCG,2mmol/L EGCG、4mmol/L EGCG groups,each groupcontained6mice.50μl highly pathogenic Escherichia coli with the concertration of1x108cfu/ml was slowly perfused into the bladder transurethrally,established bladder infection models,after48h,perfused an equal volume (50μl) of PBS buffer, and differentconcentrations of EGCG sterile solution into the bladders of mice of corresponding groups.HE staining and immunofluorescence staining of bladder tissue were performed to observethe injury of bladder epithelium and the expression level of inflammatory factorInterleukin-6(IL-6) and granulocyte colony-stimulating factor (G-CSF) as well as assessthe prevention effect of EGCG in Escherichia coli induced acute bacterial infection in mice.Results: the degree of injury and the expression level of inflammatory factors weredecreased in the PBS,1mmol/EGCG,2mmol/L EGCG,4mmol/L EGCG groupsconsecutively.Conclusions: EGCG has effective antibacterial effect and can reduce theinflammatory response due to E. coli infection, EGCG has no negative impact or a slightnegative impact on the repair of urinary bladder epithelium.
