Abstract

objective:(1)To make SD rats postoperative intestinaladhesion model, for exploreing the relation between dopamine andpostoperative peritoneal adhesion，by useing immunohistochemistry todetect the express of the Tyrosine Hydroxylase (TH) and DopamineDecarboxylase （DDC）， and Dopamine D2Receptor （DAD2R）at cecum, in the2nd,5th and8th days after operation.(2) To observe theinterreaction between dopamine (DA) and inflammatory mediator, byseparately inhibiting dopamine system and inflammatory reaction in SDrat of intestinal adhesion model, and discuss primarily the mechanism ofthe dopamine’s effect on intestinal adhesion form. Methodes：(1)Ninety-six SD rats were randomly divided to three groups: normal controlgroup（n=14），sham group （n=42）and postoperative adhesion modelgroup （n=42）.Normal control group ware done nothing, Group shamonly underwent on-off of abdomen, adhesions model were induced byEllis’ path. The rats of the latter two groups were executed in batches at2nd,5th and8th days after operation，each group had14rats wereexecuted at a time. The first group rats were executed at after feeding3days.The all executed animal were fasted12hours,at same time, theircecum were replaced after executed, in addition, the fixed preparation andparaffin-embedded specimen processed with immunohistochemistry. IPP5.1software were used for image analysis，the mean of opticaldensity value (OD) were on behalf of the TH、DDC、DAD2R in eachgroup expression. The spss17.0software was applied for statisticalanalysis, and the differences among the three groups were compared withsingle factor analysis of variance，they were statistically significant asP<0.05.(2) there were forty-two SD male rats were randomly dividedinto three groups: group A:normal saline group(n=14) and groupB:domperidone group(n=14)， group C:phenylbutazone group(n=14)，allrats were induced to adhesions model by Ellis’path.All groups were givedrelevant drug through gavage from the second day post operative, Thethree of them are respective1ml of0.9%normal saline,1ml of0.09%domperidone (Clinical equivalent dose) and1ml of0.94%phenylbutazone (Clinical equivalent dose). The blood samples of allgroups were collected after narcotized，at the same time，animal cecumwere replaced on the fifth day after surgery. The cecum were rankedmacroscopic observation,and the express of tyrosine Hydroxylase (TH)and dopamine decarboxylase（DDC）,dopamine D2receptor （DAD2R）were detected by immunohistochemistry. In addition, The levels oftransforming growth factor-β1(TGF-β1) and tumor necrosis factor-α（TNF-α），prostaglandin E2(PGE2) were determined by enzyme linkedimmunosorbent assay，count of white blood cell (WBC) were measured.The image and the data processing just as the first part experiment.Results:(1)Compared with group sham and normal control， the OD value of TH，DDC and DAD2R were all improved obviously at at2,5and8days after surgery（p<0.05、p<0.01）. In model group, the OD valueof TH，DDC and DAD2R were maximum after the second day postoperative，and the express of the eighth day were basic close to the othertwo groups，they were a gradual decline process.(2) The levels ofTGF-β1, TNF-α, PGE2, WBC in domperidone group were lower thannormal saline ones(P<0.05), at the same time the HE dyeing result showsthe corresponding change, in addtion, the express of TH, DDC andDAD2R in phenylbutazone group were lower than normal salineones(P<0.05). Conclusion：(1) The TH, DDC and DAD2R wereincreased in postoperative intestinal adhesion form process，that implythe dopamine is one of the reasons that influence the formation ofpostoperative intestinal adhesion in the early stage after operation.(2)The inflammatory response could affect gastrointestinal motion throughincreasing the express dopamine and its receptor system in the process ofintestinal adhesion formation. Meanwhile, the dopamine system couldalso aggravate the inflammatory, They influence each other and jointlyparticipate in the formation of postoperative intestinal adhesion.
