Objective:To study the correlation of HMGB1and autophagy signal Beclinl in acute pancreatitis injury. for producing a new theoretical foundation to the development of acute pancreatits about the clinical treatment and the mechanism.Methods:(1)44SD rats (350±30g) were randomized to sham operation group (SO)(n=12) and acute necrotizing pancreatitis group (ANP)(n=32). The experimental group was retrograde injected of5%sodium taurocholate into the pancreatic duct to produce the model of acute necrotizing pancreatitis. Rats were killed at3h,6h,12h and24h after sodium taurocholate injection by heart blood letting after anesthesia.(2) The levels of amylzyme and inflammatory factor such as HMGB1, TNF-alpha, IL-1and IL-6in peripheral blood were detected by Elisa assay.(3) The pathological of pancreatic tissue were observed by H&E staining. Immunohistochemistry assay used to detect the expression levels of HMGB1and Beclinl in pancreatic tissue, and then, IHC results were quantitatively analyzed by software.Results:(1) The acute pancreatitis modle was made successfully, levels of amylase were significantly increased than SO group (p<0.05), The expression levels of early inflammatory factor such as TNF-α, IL-1and IL-6in ANP group were significantly higher than SO group (p<0.05), reached the peak at6h. The HMGB1expression levels did not increased obviously at3h, but significantly increased at6h, and continued to increase with the passage of time.(2) IHC and quantitative analysis showed that the expression levels of HMGB1in ANP group were beginning to increase at6h, significantly increased at12h and continuing stay higher at24h (p<0.05). The levels of Beclin1significantly higher in ANP group with the development of the inflamation (p<0.05).Conclusion:(1) As a late inflammatory factor, the expression of HMGB1in the acute pancreatitis was obviously later than the early inflammatory factor IL-1, TNF-a, and IL-6. HMGB1may play a key role in maintaining the development of the acute pancreatitis.(2) HMGB1may maintain and promote autophagy signal Beclin1expression, thereby aggravated the development of the pancreatitis.
