Mandibular Defect Repaired by HGF Combined with PluronicF-127Gel

Abstract


Objective The present study was designed to evaluate whether the compound of hepatocyte growth factor (HGF) and Pluronic F-127gel are associated with fixed direct recrument of bone marrow mesenchymal stem cells (BMSCs), and the healing process of bone defects.Method Material of HGF-gel was prepared first. Transwell chamber was used to observe the migration of BMSCs towards HGF-gel. The cells that travel through the filter membrane of Transwell chamber were counted under the microscope of400times. Round bone defects of3mm diameter were established in30SD rats’bilateral mandibuls. In the right side of mandible defects, HGF-gel was injected. The other side of defects were compared group. After that, the wounds were sutured and each rat was injected antibiotic for prevention of infection. The animals were randomly divided into5groups and sacrificed on the1st,2nd,4th,6th,8th weeks after surgeries. Observed the bilateral bone defects by dental X-ray set. Used histochemical method of bone gla protein (BGP) and CD34to detect the growth of new vessels and the formation of new bones in bilateral bone defects. The sections of BGP were analysed by Image-Pro Plus, a software of cells’ colour image analysis. Then the images were graded by immunohistochemical score. New vessels’numbers of the sections of CD34were count under the microscope of400times. All the datas were analysed by t-test through the statistic software of spass19.0.Result Fixed direct recruitment of BMSCs in HGF-gel group was more than the group of controls (P<0.05). Wounds of the animals’bilateral mandibular were primary healing after surgeries. The bone defects of HGF-gel sides repaired faster than the control sides in every time points. Materials were absorbed since the6th week after surgeries. Through X-ray, the size of bone defects in HGF-gel group were small than the control group, as well as the bone density of HGF-gel group was better than control group. Analysed the data of the immunoreactive scores and the new vessels’numbers, it was found that the formation of new bones of HGF-gel sides were active than control sides (P<0.05) in the1st,2nd,4th,6th weeks after surgeries, and no obvious distinction (P>0.05) in the8th week, but the color intensity was declined in the8th week. In addition, HGF-gel sides has more new vessels than the control sides in the1st,2nd,4th,6th weeks (P<0.05) after surgeries, and no obvious distinction (P>0.05) in the8th week, but the vessels numbers were declined from the6th and8th week.Conclusion HGF-gel may accelerate BMSCs migration, furthermore it can enhance new vessels’growth and new bones’ formation.
