Since Hursey and Malette discovered that zeolite and chitosan were excellent hemostatic in1980s, respectively, more and more attention have paid to their coagulant activity. Meanwhile, with the understanding of interaction between solid materials and protein developed, the hemostatic interacting with plasma protein have aroused more interest. In addition, the influence factors have been investigated in order to develope an excellent performance hemostatic material.On the basis of our previous studies, this thesis further proposes that the surface hydroxyl groups of zeolite play an important role in its coagulant activity. The reduction of the surface hydroxyl groups content after calcination is characterized by TG-DSC, in situ FT-IR and XPS. The lower content of surface hydroxyl groups on the surface of zeolite leads to a stronger interaction between zeolite and plasma proteins, resulting better procoagulant performance.For further exploring the interaction between hemostatic and plasma protein, this thesis studies the chitin and chitosan materials, additionally. Accoarding to Western-blot, thrombin which is in-situ generated in plamsa can been stabilized on the surface of chitin and chitosan. The stability performance of chitosan-plasma complex is demonstrated by thrombin chromogenic substrate assays and in vitro clotting assays. In addition, the degree of deacetylation of chitosan influences the chitosan-plasma complex procoagulant performance.
