Abstract


The duck plague Anatipestifer disease (commonly known as duck infectious serositis ) by Riemerella silent Bordetella (Riemerella anatipestifer, RA) cause of ducks, geese , turkeys and a variety of other birds of a highly pathogenic contagious disease . 21 RA serotype has been reported , the lack of cross-protection between serotypes , vaccine-preventable disease brought inconvenience . The vast majority of duck the domestic duck centralized region occurred RA infection , and there is the problem of persistent infection , its mechanism is unknown so far . In addition, the study of the the RA pathogenesis of virulence factors remain blank . In this study, through the preparation of serotype 1 , type 2 RA monoclonal antibodies to detect cross-reactivity with RA different serotype isolates , aimed at finding common antigen between serotypes RA for immunodiagnostic and vaccine development . In this study, a total of 14 monoclonal antibodies against serotype 1 RA and 12 monoclonal antibodies against serotype 2 RA . ELISA tests showed 14 serotype 1 monoclonal antibody of the same serotype 1 strain and serotype 6 , 12 and 15 type strains were reactive common epitope between the serum 1,6,12 and 15 type RA bit. 12 serotype 2 mAb in the three strains with all been detected in RA has reactive , indicating that the the different serotypes RA strains having a common epitope . Biofilm formation is usually associated with the virulence of the bacteria . This test measures the ability of the 43 RA biofilm formation , and measured CH3 strains to form biofilms biochemical characteristics change with different biofilm forming ability the 3 RA ducklings isolates pathogenic . The results show that RA strains were able to form a biofilm ( 18 / 43 ) , glucose can inhibit biofilm formation but sucrose had no effect on biofilm formation , biofilm formation enhanced RA resistance to antibiotics and detergents . RA biofilm forming ability of pathogenic no direct correlation , but the long-term survival of environmental resistance enhancement enables the formation of biofilm bacteria , and perhaps one of the reasons duck RA infection continued existence . In this study, the use of a suicide vector for the first time successfully constructed RA Th4 OmpA deletion mutant strains , and to lay the foundation for further study of the role of OmpA in the pathogenesis of RA .
