Abstract


The chemical constituents from the the roots and stems of Cerbera manghas were studied, and the chemical constituents and their physiological activities from the genus Cerbera were reviewed in this thesis.1. The antitumor activities and the antioxidant activity of the extracts and the compounds from C. manghas were selected through MTT assay and DPPH free radical scavenging method in order to screen bioactive agents of medicinal resources from mangrove. Bioassay-guided fractionation led to the isolation of 20 compounds from the ethanol extract of the roots and stems of C. manghas by column chromatography, and the structures of 8 compounds were indentified by physicochemical and spectral data (UV, IR, MS, 1D NMR, and 2D NMR) as two cardiac glycosides, (-)-17β-neriifolin (1), and 17β-Digitoxigenin-β-D-glucosyl-(1→4)-α-L-thevetoside (2), two iridoids, Cerbinal (3), Coniferaldehyde (4), one organic Acid, Vanillic acid (5), and two lignans, (-)-olivil (6) and (+)-Cycloolivil (7), 8-Hydroxypinoresinol (8). Compounds 5 and 8 were isolated from this genus for the first time.2. The chemical constituents from the petroleum ether fraction of the ethanol extract from the roots and the stems of C. manghas were analyzed by gas chromatography-mass spectrometry respectively. Twenty-four compounds were identified from the petroleum ethanol fraction of the roots of C. manghas. Twenty-eight compounds were identified from the petroleum ether fraction of the stems of C. manghas.3. The antitumor activities of the extracts and the compounds from C. manghas were tested by MTT assay. Both the ethanol extract and the methylanol extract exhibited strong cylotoxicities, and the methylanol extract showed significant cytotoxicities. Among the isolated compounds, the three cardiac glycosides exhibited strong inhibitory activity towards chronic myelogenous leukemia (K562), human gastric carcinoma (SGC-7901), and human hepatoma (SMMC) cell lines.4. The antioxidant activity of the extracts and the compounds from C. manghas were tested by DPPH free radical scavenging method. Among the extracts, the ethanol extract and the methylanol extract from the roots of C. manghas showed antioxidant activity, while the extracts from the stems exhibited lower antioxidant activity. The three lignans showed strong antioxidant activity.
