Abstract


2l,4’-Dihydroxy-6’-methoxy-3’,5’-dimethylchalcone (DMC) is a chalcone, which is isolated from the buds of Cleistocalyx operculatus. Acute liver failure (ALF) refers to a serious disease which has witnessed the emergence of a highly lethal condition termed acute liver failure including hepatocellular death, extrahepatic complications and hepatocyte regeneration. In this paper, the hepatoprotective effects of DMC against CCl4-induced acute liver injury in Kunming mice as well as its mechanism were investigated.Firstly, we investigated the hepatoprotective effects of DMC against the cytotoxicity induced by carbon tetrachloride (CCl4) (4 mM,3 h) in L02 hepatic cells. The results showed that pretreatment with DMC significantly decreased activities of serum hepatic enzymes like alanine aminotransferase (ALT). Pretreatment of DMC was found to be comparable to the reference drug BDP.Secondly, we investigated the hepatoprotective effects of DMC against CCl4-induced acute liver injury in Kunming mice. The results showed that pretreatment with DMC significantly decreased activities of serum enzymes like ALT and aspartate aminotransferase (AST). Pretreatment of DMC was found to be comparable to the reference drug BDP.Finally, the hepatoprotective effects of DMC and its mechanism against CCl4-induced acute liver injury in Kunming mice were investigated. These results combined with liver histopathology demonstrate that DMC has potential hepatoprotective effects, which may be related to the attenuation of oxidative stress, increasing the antioxidant cascade and inhibition of lipid peroxidation.This study could serve as a useful reference to allow the future exploitation of DMC as a hepatoprotective medication for the therapy of oxidative stress-induced liver damage.
