Rigor mortis was a very important stage of biochemical changes in muscle after slaughter and it had negative impact on meat quality and processing characteristic. The speed and intensity of rigor mortis will affect the course of meat ageing and meat quality. How to shorten the course of rigor mortis and reduce the intensity of rigor mortis in order to improve meat quality became a important technical issue. Because the course of rigor mortis had closely correlation with energy metabolism and limiting enzyme of glycolysis, while there was little study on adenylic acid degradation and limiting enzyme of glycolysis in rigor mortis of three-yellow chicken after slaughter, it was necessary to study energy metabolism rules in rigor mortis of chicken breast muscle by different fasting treatment and different temperature treatment and research the correlation between limiting enzyme activities of glycolysis and the content of energy substance in order to provide evidence for regulating the course of rigor mortis and illustrating the nature of rigor mortis.In the first part we studied the impact on the energy metabolism of rigor mortis of breast muscle after three-yellow chickens were slaughted by no fasting treatment, 12 hours fasting and 24 hours fasting. Chickens breast muscle were taken out quickly after slaughter for different fasting treatment groupes, chicken breast muscle was put into liquid nitrogen one time at every 15 minutes from 0 to 3 hours at 15℃, these samples were used to measure all kinds of indicators. The content of ATP、ADP、AMP and IMP in breast muscle of rigor mortis was measured by HPLC. The activities of related enzymes were measured by kits in the course of rigor mortis. The results indicated that at 15℃the content of ATP in breast muscle decreased gradually in 3 hours after slaughter on the whole for the different fasting treatment groups. At 15℃, the content of AMP in breast muscle increased at first, and then decreased in 3 hours after slaughter. At 15℃, the content of IMP in breast muscle increased at first, and then a little decreased in 3 hours after slaughter on the whole. The activities of HK in breast muscle increased at first, then decreased. The activities of LDH in breast muscle decreased in 3 hours after slaughter at 15℃, and the result indicated that there was great impact on the change of PK activity in rigor mortis of chicken breast muscle by fasting treatments.In the second part we studied the impact on the energy metabolism of rigor mortis of chicken breast muscle by different ambient temperature treatment groupes after slaughter. Chickens breast muscle were taken out quickly after slaughter and put in the environment at 15℃and 4℃respectively. Chicken breast muscle was put into liquid nitrogen one time at every 30 minutes from 0 to 4 hours at 15℃and 4℃, which was used to measure all kinds of indicators. The content of ATP in breast muscle decreased gradually in 4 hours after slaughter on the whole for the different ambient temperature treatment groups. The content of AMP in breast muscle increased at first, and then decreased. The content of IMP in breast muscle presented increasing trend. There was great impact on the change of content of ATP、ADP、AMP and IMP in chicken breast muscle in 4 hours after slaughter by different temperature treatment.The activities of HK in breast muscle increased at first, then decreased trend in 4 hours after slaughter by different temperature treatment groupes. The activities of LDH in breast muscle decreased in 4 hours after slaughter. The result indicated that there was great impact on the change of HK and PK activities in rigor mortis by different ambient temperature treatments after chickens slaughted.This study indicated that there was important impact on energy metabolism during rigor mortis of chicken breast muscle by different fasting treatments and different ambient temperature treatments after slaughter.
