Abstract


To investigate the adaptive formula of fermentation medium of three kinds of edible and medicinal mushrooms by utilizing mycelium as the target product,the Coprinus comatus,Coriolus versicolor and Schizophyllum commne Fr,which collected and domesticated from Chinese Academy of Agricultural Sciences Institute of Agricultural Resources and Regional Planning were employed as the experimental materials.Different types of extracting rate of polysaccharides were determined through extracting the polysaccharide of mycelium and fermentation liquid.To investigate the adaptive technique of polysaccharides extracting,the mycelium and fermentation liquid of Coriolus versicolor were employed as the experimental materials.Furthermore,Lewis lung cancer solid tumors were used to investigate the influencing of polysaccharides to the tumor inhibitory rate,in order to exploring the function of different kinds polysaccharides.This can establish scientific basis for the exploring of wild edible and medicinal fungi.Main results are as follows:1.In the appropriate cultural environment,the excellent formula liquid cultural medium for the fungi growth and mycelium production were selected by the comparative experiment of cultural medium formula of edible and medicinal fungi.①The optimum liquid medium of C.comatus.Sucrose 30g,Corn Flour 20g,Bran 40g, KH2PO41g,MgSO40.5g,H2O 1000ml。②The optimum liquid medium of C.versicolor:Potato 200g,Sucrose 20g,KH2PO4 1g, (NH42SO4 1g,H2O 1000ml。③The optimum liquid medium of S.commune.Potato 200g,Bran 100g,Glucose 30g,Yeast extract 1g,KH2PO45g,MgSO42.5g,VB10.1g,CaCO33g,H2O 1000ml。2.The polysaccharide extraction of three kinds of mycelium and fermentation liquid were investigated.The results indicated that there were significant differences of the yield rate among distinct mycelium and fermentation liquid by the same extracting method.The yield rate of polysaccharide from mycelium of C.comatus was 16.5%,was highest;and the polysaccharide from fermentation liquid of S.commune was 65.0%,was highest. 3.The polysaccharide content and protein content of three edible and medicinal fungi were determined.The results shows that the seriatim of the mycelium polysaccharide content is C.comatus>S.commune>C.versicolor,and the seriatim of the polysaccharide content in Polysaccharide from fermentation liquid was the same.4.The adaptive technique of mycelium polysaccharides extracting of S.commune is:freeze drying,ultrasonic time 10 min,ultrasonic power 600W,solid-liquid ratio 1:30,extraction time 4 h. The adaptive technique of fermentation liquid polysaccharides extracting of S.commune is: ultrasonic time 10 min,ultrasonic power 600W,alcohol precipitation time 8 h,alcohol precipitation ratio 1:5.5.The inhibition function of three types of medical mycelium to Lewis lung cancer was studied. The results indicated that all of the three types of mycelium polysaccharide can inhibit the tumor. The inhibition ratio of C.versicolor polysaccharide to Lewis lung cancer is highest,which reached to 86.8%.
