Rotigotine, a potent and selective D3/D2/D1 dopaminergic receptor agonist, has direct impacts on these receptors, relieve tremor and movement disorders, reduced muscle tension, therefor, achieve similar treatment as dopamine. Its pharmacodynamics is better than apomorphine, bromocriptine, and pergolide. Foreign clinical results show that rotigotine is safe tolerance and effective drug for the early period of Parkinson’s disease,and for later period Parkinson’s disease ,it can be a supplementary drug of L-dopamine.However, rotigotine has extensive first-pass metabolism,short half-time, very low oral bioavailability, making the per oral delivery route not feasible. but it has high-fat-soluble,that make it is suitable for transdermal system.In this thesis, some physicochemical properties were studied, the system of rotigotine iontophoresis transdermal was confirmde , some factors to the drug transdermal flux in vitro werestudyed, such as current density , duty cycle of frequency ,drug concentration, penetration agent, pretreatment of skin, competitive ions, The best conditions of iontophoresis wes selected out. The best experimental conditions as follows:the donor liquid was citric acid buffer salt solution which pH was 5.0(the concentration of concentration was 0.07M),the acceptor liquid was phosphate buffer solution which pH was 7.4,pulse current was employed, current density was 0.5mA/cm2, duty cycle of frequency was 1:2, Drug concentration was 1.4 mg/mL. Under the best experimental conditions ,the iontophoretic flux of rotigotine hydrochloride was 93.74μg/ cm2·h-1 .,which is 11.07 times of the foreign reference substance.The method of HPLC has been developed to determine the content and relative substances of rotigotine. Homogeneity, non-aqueous titration of rotigotine content was also developed.The results above studies indicate that iontophoresis was a new, effective penetration method for transdermal. Hence,rotigotine iontophoresis transdermal is a desirable topics, which has good prospects for the development and research space.
