Antiapoptosis Gene, Livin Expression in Endometrial Carcinoma and Its Correlation with Cell Proliferation and Apoptosis

Abstract


Objective To detect the expression levels of Livin in endometrial carcinoma and normal endometrial tissues. Their association with clinicopathological characteristics of endometrial carcinoma was then analyzed. We explored the relationship of Livin expression and their effects on proliferation and apoptosis of endometrial carcinoma cells, to discuss their roles in the genisis and development of endometrial carcinoma.Methods Immunohistochemical SP, RT-PCR and Western-blot method were used to detect the expressions of Livin in 35 cases of endometrial carcinomas and 30 cases of normal endometrial tissues. Flow cytometry was used to detect proliferation and apoptosis of carcinoma cells.Results 1. The expression of Livin gene: Immunohistochemical SP method showed that the positive rate of Livin gene was 62.86% in 35 endometrial carcinoma tissues, but Livin gene was not expressed in 30 normal endometrium tissues. The RT-PCR and Western-blot methods showed that the positive rate of Livin gene was 68.57% in 35 endometrial carcinoma tissues, but Livin gene was not expressed in 30 normal endometrium tissues. The expression of both variants was simultaneous basically. The RT-PCR and Western-blot methods have identical results. 2. Correlation between expression of Livin gene and clinicopathological factors: There was no significant correlation between positive Livin gene expression and age, histological types, differentiation grades, infiltration degree and lymph node metastasis (P> 0.05). The expression of Livin gene was closely related to pathological staging (P<0.05). 3. Correlation between the expression of Livin gene and tumor cell proliferation in endometrial carcinoma tissues: The average proliferation ratio in 35 endometrial carcinoma tissues was (29.42±5.58) %. The average PI of 24 cases with positive Livin expression and 11 cases with negative expression were respectively (31.55±6.19) % and (27.00±4.26) %, with significant deference (t=2.598, P<0.05). 4. Correlation between the expression of Livin gene and cell apoptosis in endometrial carcinoma tissues: The average ratio of early apoptosis in 35 endometrial carcinoma tissues was (1.87±0.29) %. The average apoptosis ratio of 24 cases with positive Livin expression and 11 cases with negvative expression were respectively (1.79±0.25)% and (1.96±0.37)%, with significant deference (t=2.146, P<0.05).Conclusion 1. Livin gene is highly expressed in endometrial carcinoma tissues, but shows no expression in normal endometrium tissues. 2. There was no significant correlation between Livin gene expression and age, histological types, differentiation grades, infiltration degree and lymph node metastasis, but the expression of Livin gene was closely related to pathological staging. 3. The expression of Livin gene in endometrial carcinoma tissues was correlated with tumor cell proliferation. 4. The expression of Livin gene in endometrial carcinoma tissues had correlation with cell apoptosis.
