Objective: To observe the the lumbosacral dominant spina bifida fetal rat gluteal muscle motor end plate and muscle development, morphological changes, and the possibility of transplantation to provide theoretical reference for the lumbosacral dominant spina bifida gluteal muscle. : Modeling: pregnant 9d healthy Kunming mice were randomly divided into two parallel groups (5), and the control group (5) and the experimental group (15). In to pregnancy 9d0h given experimental group of pregnant rats three times continuously fed with all-trans retinoic acid, dose: 3 × 10ml/kg.bw medication time turn interval 6h. The control group of the same Act gavage doses of soybean oil, parallel-group normal feeding groups were pregnant 18 days to remove the fetal rat caesarean: all-trans retinoic acid fed group shared the fetal rat 123, which lumbosacral skin integrity dominant spina bifida fetal rat 70, the lumbosacral skin defect explicit spina bifida fetal rat 16, the appearance of normal fetal rat 23 other abnormal fetal rat 14; parallel groups of pregnant rats were obtained 51 fetal mice in the control group pregnant rats was 44 fetal rat, parallel-group, and the appearance of the control group showed no abnormal fetal rat. Research content OUTCOME MEASURES: gluteus morphological pathological changes (gross and HE staining); gluteal muscle motor endplate zone distribution and morphology stained with acetylcholinesterase staining, the professional image processing software to calculate the average light motor endplate zone density and integrated optical density values. Statistical analysis: Numerical analysis of variance, using the statistical software for data processing, P <0.05 indicates statistically significant difference. Results: 1. Morphological observation: general observation see fetal rat gluteus start and end surface, diamond-shaped, top-down from the outside and sacrum on the back of the iliac wing muscle bundle under oblique and short tendon from the iliac after the ligament between the spine, buttocks after line after iliac hip surface, iliac lower coccyx and two bone ends with intertrochanteric. Groups fetal rat gluteus appearance of red, look no big difference; HE staining under light microscope: D 1 Group D 2 group the D 3 < / sub> were observed in the spinal canal and spinal cord structures complete spinal cord gray matter clear demarcation, the central canal of the middle, can see the complete distribution of neurons and glial cells, high magnification can also be found in neurons and glial cytoplasmic Nissl bodies. Gluteus muscle fibers clearly obvious stripes, normal muscle fiber morphology; T 1 group fetal rat lumbosacral spinal cord tissue edema, increased interstitial cell bodies of neurons slightly round, lightly stained cytoplasm, nerve yuan less than the other groups, the number of glial cells legible. Fetal rat gluteus muscle fibers arranged rules clear and obvious stripes; T 2 group fetal rat lumbosacral spinal cord tissue edema, loose organizational structure, messy, significantly increased interstitial few neurons gliosis, scattered red neurons. Gluteus muscle fibers arranged slightly disordered, clearer obvious stripes; gluteal muscle movement endplate distribution and speciation: D 1 Group D 2 Group D 3 group gluteal muscle motor endplate is located in the the the gluteal muscle deep surface or intermediate Ministry was \T 1 group and T 2 group gluteal muscle motor endplate located gluteus deep or middle portion, was the \ group motor endplate staining than D 1 Group D 2 Group D 3 gluteal muscle motor endplate staining shallow, light stained uniform; T 2 group gluteal muscle motor end plate in addition to staining than D 1 group D 2 group the D 3 < / sub> group motor endplate staining shallow, short, dyed uneven brown color, accompanied by the motor endplate deformity or defect; 3.T 1 group motor endplate average optical density ( 0.8500 ± 0.0310), and integrated optical density values ??below the D 1 group (0.8625 ± 0.0248,8200.0330 ± 235.8933) (8079.4296 ± 102.4552), D 2 group (0.8574 ± 0.0144 , 8145.6800 ± 136.5118), D 3 group (.8691 ± 0.0153,8255.9750 ± 145.8148): T 2 group motor endplate average optical density (0.8124 ± 0.043) and integrated optical density value (7718.5600 ± 413.7121) below T 1 group, D 1 Group D 2 group, D 3 sub > group average optical density and integral optical density value; 4. statistically analyzed using the statistical variance analysis, T 1 group average optical density and integral optical density value D of 1 group, 2 group, D 3 group, no significant difference (p> 0.05); T 2 group average optical density and integral optical density value D of 1 group D 2 group, D 3 group optical density and integral optical density value, there is a statistically significant difference significant statistical significance (p <0.01); T 1 group, the average optical density and integrated optical density comparison with T 2 group, the average optical density and integral optical density values, the difference significance (p <0.01).
