Abstract


The establishment of the blood-testis barrier and spermatogenesis are inseparable. The blood-testis barrier is used to protect the reproductive cells in the testis not own immune system and blood the harmful molecules injury physiological barrier , continuous tight junctions between cells composed of the adjacent support . Closely linked to form a natural barrier that can prevent proteins and other macromolecules by creating a good environment for germ cell differentiation and development . Occludin and claudin-11 is the major mammalian blood-testis barrier tight junction protein . This study to explore the expression of these two proteins in amphibian testis African clawed frog (Xenopus laevis) study , using RT-PCR , Western blot and immunofluorescence method to confirm the occludin and claudin-11 in Xenopus testis expression ; the mature Xenopus testis before and after occludin and claudin-11 mRNA expression by Real-time PCR quantitative analysis and comparison . The results show that mature around the testis of Xenopus occludin and claudin-11 had higher levels of expression , but no significant differences in mRNA expression levels . The experimental results with the expected substantially match, indicating that the occludin and claudin-11 are two stable and high expression of the protein in Xenopus testis . These two proteins may be similar to the function of the mammalian blood-testis barrier , to provide a safe environment for Xenopus spermatogenesis , sperm normal formation . This study may provide first-hand information to further explore amphibians the existence of the blood-testis barrier structure .
