Abstract


Liposome is a kind of closed vesicle with bilayer structure similar to biomembrane, which is the most convenient and safety temperature-controlled release drug carrier. Liposome composed of phospholipids and cholesterol, and cholesterol effect on the liposome physic-chemical properties mostly. The effect of cholesterol on the structure and properties of the liposome has always been the focus recently.In this work, we focus on the liposome membrane structure as a function of cholesterol concentration and the effect of the membrane structure change on the liposome properties and function. A monodisperse berberine liposome was prepared by supercritical CO2 method. Fluorescence intensity and anisotropy of fluorescence probers in the liposome were measured using fluorescence methods, and the effects of cholesterol concentration on the fluorescence intensity and anisotropy were studied, and the fluidity and microviscosity were obtained for the liposome bilayer membrane. The relationships between the liposome membrane microstructure and the physical properties in the liposome with various cholesterol concentrations were discussed. Berberine hydrochloride used as a model drug was entrapped in the liposome, and the effects of cholesterol concentration in the liposome on drug entrapment efficiency and control release were revealed.The results were shown that the microviscosity increases with cholesterol concentration, and the fluidity decreases with increasing cholesterol concentration for the liposome bilayer membrane when cholesterol concentration is below 40%. The similar result was obtained from the micropolarity of liposome membrane bilayer studied by pyrene probe. The size, zeta potential, lower critical solution temperature (LCST) and entrapment efficiency of liposome also increase with cholesterol concentration under the cholesterol concentration of 40%.
