Abstract


The Chinese Yam is a medicine-food efficient characteristic economic crops, because of these varieties have complex genetic background, the identification and classification is difficult by morphological characteristics . The ISSR molecular marker is one of practical and effective method to identified the germplasm of yam, its can provides the proof and technical support at molecular level for the choosing and using of parents and construction of core collection of yam breeding. Therefore, in this paper ,analysis of genetic diversity of the 94 germplasms yam from 13 different provinces of China were studied by the ISSR molecular marker, and the agronomical traits of germplasm of yam were investigated and classified, The main results of the study were as follows:1. Study on Morphological Diversity on yam (1)The blade length were between of 2.95 to 15.07㎝. The Wingedyam were between of 10 to 15㎝, The Dioscorea opposite Thunb were between of 6 to 10㎝ , The Rhizoma Dioscorea persimilis were between of 5 to 10㎝, Yam were between 3-8㎝. The blade length were sm41 minimal, sm47 maximal. (2)The leaf width were between of 2.72-12.31㎝. The Wingedyam were between of 6-12㎝, The Dioscorea opposite Thunb were between of 4 to 7㎝ , The Rhizoma Dioscorea persimilis were between of 4 to 8㎝, Yam were between 3-7㎝. The leaf width were sm41 minimal, sm57 maximal.(3)The leaf index were relatively stable, between of 1.0-1.7㎝. The Wingedyam were between of 1.1 to1.7㎝, The Dioscorea opposite Thunb were between of 1.4 to 1.7㎝ , The Rhizoma Dioscorea persimilis were between of 1.0 to 1.6㎝, Yam were between 1.1 to 1.6㎝. (4)The petiole length were between of 1.34 to 9.54㎝, sm41 minimal, sm46 maximal.(5)The the content of dry matter were between of 6.85-42.65%.2. Study on genetic diversity by ISSR: ISSR polymorphisms of 94 varieties of germplasms yam were studied by PCR with 24 different primers which Selected from 100 random primers. 356 loci were amplified, of which 342 (96.6%) were polymorphic. 11~22 loci were amplified in every primers, 14 loci mean. There were maximum number of 22 loci of I40 among these primers. Then Clustering analysis t of UPGMA by NTSYS 2.10e , The results showed that:when the similar coefficient was up to 0.74, The 94 varieties of germplasms yam could be sorted into four species , sm1、11、15、33、97 ect. 20 yam belonged to I type;sm14、23、54、65 ect. 20 yam belonged to II type (Dioscorea alata L) ; sm12、39、43、44、69-1、76、79 ect. 7 yam belonged to III type (Dioscorea opposite Thunb ); sm5、22、87、89 ect. 43 yam belonged to IV type (Rhizoma Dioscorea persimilis). These conclusions are basic with the classical classification of yam morphology of Peijian.
