Abstract


Plants affected by canker, will host a series of plant disease resistance-related reactions, including plant response to the pathogen invasion, changes in antioxidant enzymes, these reactions poplar clones at different spatial and temporal differences bigger. Since the 1980s research has achieved many significant results, but in the production of the main varieties of poplar resistance identification field combining indoor Resistance Identification less. In this thesis, poplar canker resistance on the theme, with 69 poplar, Populus Shangzhou, 84K Yang, Shaanxi Forest on the 4th Young, E24-01 Young, in Hua Changqing Yang, 108 Young, 107 Young, South anti- Young, J2 Young, Nakabayashi Mei Ho Young, No. 12 in Sui, the black defense, Evolution and Young, North anti-Young, 2001 Young cultivars poplar species as the research object to callus, in vitro tests for the interior branches materials, combined with the resistance of experimental forest surveys; apply this test in vitro resistance test shoots inoculation, groups of tissue culture, physiological and biochemical techniques and methods of production of the main varieties of poplar canker resistance. Detected after vaccination canker antioxidant enzymes (SOD, CAT, POD, APX) activity changes, changes in the analysis of these enzymes resistant and susceptible poplar difference between; observed canker infection in the host process, and to explore the peroxidase role in the process of plant disease resistance. Preliminary study of the main varieties of poplar and canker bacteria produce an allergic reaction, and strive to reveal multiple perspectives on poplar canker resistance mechanisms. The main results are as follows: 1. Production for 16 major varieties of poplar Experimental Forest consecutive years of ulcer disease investigation, the calculation of the various poplar clones disease index, annual differences in resistance. The results showed that: After the young poplar plantation substantial morbidity, planting time most poplar clones enhanced disease resistance, resistance to different poplar clones are quite different from year to year due to differences in the resistance of the performance of different varieties varying the resistance of which 107 Young influenced by rainfall, rainfall increases with the decrease. The results showed that: when vaccination canker 12h 16 varieties callus Jiewei observed canker, 24h there mycelium callus extended to different factions of hyphae invade poplar speed performance vary greatly, different poplar Clones hyphae at different speeds. Shangzhou at 48h 80% Populus callus have all been covered by mycelium, hyphae more sparse, all varieties were inoculated at a slight browning. Callus inoculated ulcer activity of POD activity upward trend is more obvious, a greater increase in resistant varieties, peaked later. SOD, CAT activity decreased at first and then showing a clear peak, APX activity most poplar clones small changes, including changes in the biggest anti-black, Shangzhou Populus's APX activity has been in a downward trend.
