Abstract


In this thesis, the alpha-galactosidase enzyme preparation used in corn - soybean meal diets for weaned piglets performance, fecal microorganisms, blood biochemical parameters and the apparent digestibility of nutrients, and further dialysis tube in vitro digestion Method glycosidase and effect of alpha-galactosidase to be assessed. Test: In order to investigate the different levels of the enzyme preparation weaning growth performance, selected 90 healthy 37-day-old long * Large weaned piglets (weaned at 30 days of age), the average weight of 11.05 ± 1.97kg. Completely randomized design, according to the same source, the principle of initial body weight similar pigs were randomly divided into five treatments, each treatment with three replications, each repeated six pigs (female). Ⅰ was the control group, only fed corn - soybean meal basal diet, do not add the alpha-galactosidase enzyme; Ⅱ, Ⅲ, Ⅳ, Ⅴ for the test group, respectively, in the basal diet supplemented with different levels (20U / t , 30U / t, 40U / t, 50U / t) of the alpha-galactosidase. The results showed that: add different levels of alpha-galactosidase enzyme preparation in corn - soybean meal diet could improve the production performance of weaned pigs, the growth of the test group IV the best results with the control group I, experimental group II, the experimental group III, compared to the experimental group V, the average daily gain were increased by 16.13%, 9.09%, 2.86%, 2.86%, but the difference among the groups was not significant (P GT; 0.05) Test 2: further laboratory analysis of samples collected by the feeding trials. Test 3: after the pepsin - trypsin two-step method in vitro digestion parameters (enzyme concentration, pH, time) is determined, using a dialysis tube in vitro digestion in vitro digestion of feeding test diets and common soybean rating alpha-galactosidase enzyme effect. The test results show that: (1) under the conditions of this experiment, pepsin optimum concentration of 3.0mg/ml, and the optimum pH value of 1.8, the optimal reaction time of 4h; trypsin optimum concentration of 3.0mg/ml, most suitable pH value of 7.6, the optimum time of 24h. (2) alpha-galactosidase enzyme can improve corn - soybean meal diet dry matter, crude protein digestion in vitro digestibility of energy, the effect of group IV, its dry matter, crude protein, digestible energy digestibility compared control group were increased by 10.01%, 4.65% and 2.68%; alpha-galactosidase enzyme in vitro digestion is better than diet added in soybean meal, which the highest concentration of 0.3U/kg enzyme crude protein in vitro digestibility. (3) by diet digestibility in vitro and in vivo digestibility, daily gain regression analysis yields: a strong positive correlation between in vivo dry matter digestibility (Y) and in vitro dry matter digestibility (X) ( r = 0.888, P = 0.022), the regression equation: Y = 0.6077X 56.175 (R2 = 0.7888, P = 0.044 lt; 0.05, n = 5); vitro dry matter digestibility (X) and average daily gain (Y there is a strong positive correlation (r = 0.898, P = 0.019)), the regression equation: Y = 0.0113X-0.1245 (R2 = 0.8072, P = 0.038 lt; 0.05, n = 5). Digestion in vitro tests and in vivo digestion test, the results of feeding trial, three are basically the same, indicating that the in vitro enzymatic method used to predict the effect of alpha-galactosidase enzyme preparations.
