Abstract

Turnip mosaic virus is a the worldwide popular plant virus, bring serious harm to crop production; cucumber green mottle mosaic virus is a straight baculovirus, a tobacco mosaic virus, the virus infection hoist Branch crops, causing mottled leaves, plant growth and dwarf. Therefore, the establishment of a rapid and reliable detection method, to prevent the introduction of the virus to protect the safety of agricultural production in China, has a very important significance. The electrochemical immunoassay sensor selectivity, many kinds of low-cost test, regardless of the sample color, turbidity impact, the required equipment is relatively simple, fast, small, and many. Widely used in the field of medical, food analysis, industrial production, and environmental testing. In this paper, the development of electrochemical immunoassay sensor for detecting Turnip mosaic virus and cucumber green mottle mosaic virus, the main contents are as follows: (1) combined with enzyme-catalyzed silver deposition signal amplification method to build a sensitive and simple detection wu Jing mosaic virus electrochemical immunoassay sensors. Cysteamine and glutaraldehyde fixed to the surface of the gold electrode. Turnip mosaic virus antibody was then connected via glutaraldehyde to the surface of the gold electrode, and ultimately the antibodies are immobilized on the electrode surface. Turnip mosaic virus by binding with the antibody on the sample is fixed to the surface of the gold electrode. Alkaline phosphatase labeled Turnip mosaic virus antibody fixed good Turnip mosaic virus reaction, even on the electrode surface. Alkaline phosphatase catalytic substrate - ascorbic acid-2 - phosphate, the surface of the electrode decomposition of ascorbic acid, the final silver ions are reduced to metallic silver single mass, the deposition on the electrode surface, with the linear sweep voltammetry (LSV) detecting elemental silver deposited on the surface of the electrode, the amount of the detected content of silver ions and turnip mosaic virus in a linear relationship within a certain range, the turnip mosaic virus detection by the detection of silver. Enzyme catalyzed signal amplification method is combined with electrochemical detection sensitivity so that the method can obtain a lower detection limit. Optimization of experimental conditions, the sensor signal response with Turnip mosaic virus stock dilution showed a good linear relationship in the range of 10 to 1000-fold dilution, the detection limit of 500000 times Turnip mosaic virus stock dilution. This method is simple, low cost Turnip mosaic virus biosensor detection is expected to have a certain value. (2) electrochemical biosensor based on enzymatic silver deposition signal amplification experiment constructed electrochemical detection cucumber green mottle mosaic virus immune sensor. To the CGMMV the antibody was immobilized on the electrode surface let electrodes sequentially in CGMMV to be measured, the antibody, alkaline phosphatase enzyme-labeled CGMMV incubated. Eventually, with a substrate solution in the present enzymatic silver deposition reaction. Electrochemical signal detection by linear sweep voltammetry (LSV). The method is sensitive, simple, good selectivity, wide linear range, low detection limit. Therefore, in CGMMV detection prevention expected to have a good prospect.
