Abstract

Blackwater horsefly (black soldier fly, Hermetia illucens) is rich in nutrition is an important resource insects, larvae and pre-pupae can partially replace soybean meal for pigs, chickens and tilapia aquaculture, and partial substitution of fish meal and fish oil for rainbow trout breeding. Blackwater horsefly can reduce the accumulation of feces and odors generated also able to control the breeding of houseflies, and reduce the number of E. coli in the feces. This thesis explores the biological characteristics of the different water content diet feeding Blackwater horsefly, in order to optimize the blackwater gadfly artificial breeding conditions, to improve farming techniques Blackwater horsefly. , From blackwater Horseflies surface and intestinal isolated 10 having a protein and an organic phosphorus decomposing ability of Bacillus subtilis strain, these strains with different concentrations is added to the sterilized manure blackwater horseflies biological characteristics impact of manure N, P capabilities Blackwater horsefly. The artificial feed moisture content on the biological characteristics of Blackwater horsefly research: five moisture content of feed are suitable for the growth of the Blackwater horsefly, including moisture content of 75% and 80% of the feed feeding Blackwater horsefly larvae weight, pre-pupal weight and prepupa harvest was significantly higher than the 60%, 65% and 70% moisture content, but the larvae to the the prepupae time required and the duration of eclosion significantly longer than the 65% and 70% water content. 10 having a protein and an organic phosphorus hydrolysis ability of bacteria obtained from the the blackwater horseflies larvae gut and the surface separation, wherein a strain isolated from the intestinal tract, respectively labeled S14, S15, S16, S19 and S20, isolated from surface strain. labeled T2, T4, T6, T7 and T8. Strain 16S rDNA sequences and gyrB gene (promote helicase gyrase B subunit gene) carried out molecular identification of these strains are Bacillus subtilis (Bacillus subtilis), gyrB gene sequence of these strains of B. subtilis subsp. subtilis str.168 strains and the type strain B. subtilis subsp subtilis, BCRC10255 phylogenetic tree was constructed as an internal, 10 bacteria species level obvious kinship difference and there is no duplication strain. Blackwater horsefly larvae gut separated four Bacillus subtilis S14, S15, S16 and S19, as well as a reference strain of Bacillus natto (Bacillus natto) D1 of the research carried out, in sterilized manure added at different concentrations ( 104cfu / g and 108cfu / g) of Bacillus, kept Blackwater horsefly growth and development, and chicken manure N and P utilization have a significant impact. Intestinal microorganisms added to improve the Blackwater horsefly prepupae weight, harvest pre-pupae and larvae survival rate, to improve Blackwater horsefly sterilized chicken manure N and P utilization; sterilized chicken manure to add a hay different concentrations of Bacillus the blackwater horseflies Growth no significant effect. S15 104cfu / g concentration of added processing best in all treatments. Separation utilization the blackwater Horseflies larvae surface T2, T4, T6, T7 and T8 5 strains of Bacillus subtilis Bacillus undertaken studies show that in the sterilized manure added at different concentrations (104cfu / g, 106cfu / g and 108CFU / g) Bacillus improve blackwater gadfly the prepupae harvest volume and larval survival, and be able to improve of Blackwater Horseflies sterilized chicken manure N P utilization and dry matter utilization, but Blackwater horsefly prepupae average weight, adult body length and a negative impact on life, treatment and control of the different stages of development time difference was not significant; different concentrations of the same surface microbial additive amount of the Blackwater the horsefly biological characteristics affect the outcome different. In all treatments, T2 add processing effects 104cfu / g concentration best harvest prepupae larval survival, the survival rate of larvae transformed, N, P utilization and dry matter utilization were significantly higher than CK.
