Abstract

Objective : This study by studying Pokemon on E2F-4 in HepG2 cells in the transcriptional regulation role for cancer gene therapy provide a new target . Method : Build E2F-4 expression plasmid and the different fragment length of E2F-4 promoter, liposome mediated through Pokemon expression plasmid and E2F-4 promoter , E2F-4 expression plasmid and Pokemon hepatic cancer cells transfected with the promoter HepG2 , use dual luciferase reporter gene assay after transfection E2F-4, Pokemon transcriptional activity ; real time PCR assay expression plasmid transfected Pokemon after E2F-4 mRNA expression ; chromatin immunoprecipitation technique to detect cell Pokemon inside with E2F-4 protein promoter effect relationship . Results : Pokemon expression plasmid fragments of different lengths of E2F-4 promoter was inhibited , E2F-4 expression plasmid fragments of different lengths Pokemon promoter is also inhibited . Pokemon expression plasmid transfected E2F-4 mRNA expression was significantly inhibited . Chromatin immunoprecipitation Show: intracellular Pokem-on through direct binding to E2F-4 promoter to regulate the expression of E2F-4 . Conclusion : Pokemon expression in HepG2 cells , inhibit E2F-4 promoter activity effect , Pokemon directly with E2F-4 promoter binding , can down E2F-4 mRNA levels , inhibit the expression of E2F-4 . E2F-4 in HepG2 cells were also Pokemon can inhibit oncogene promoter activity .
