Abstract

Nowadays, a large amount of antibiotics especially that with the P-diketone struature are used in the treatment and prevention of illness in human, veterinary and aquaculture, and as growth promoter in veterinary and aquaculture. Nevertheless, a great part of antibiotics and their active metabolite are excreted into the environment. The existence of antibiotics and their active metabolite in environment will probably influence the micro organization of the natural environment and lead to resistance in bacteria, and cross-resistance between human and veterinary. Hence research on the residue of antibiotics in environment and their environmental risk and to establish a sensitive and rapid pharmaceutical determination method is of great importance in environmental treatment and ecological protection. In the first part, the applications of some separation and analysis methods of antibiotics in environmental samples for the detection emerged in recent years were summarized in detail. The synthesis and application of molecularly imprinted polymer were also reviewed. In the second part, we chose ITA as functional monomer for it could strongly coordinate with metal ions. We successfully prepared the MIP using the ITA-Fe3+-CIP complex which can absorb all antibiotics that withβ-diketone structure. In this research, we took cyclic voltammetry, dynamic absorption, equilibrium absorption to identify the MIP could absorb both of the template and other molecules withβ-diketone structure and to confirm the best prepare condition. Varies preparation conditions were investigated for they are the key factor for adsorption capacity. The MIP was prepared when the ITA-Fe3+-CIP ratio and the proportion of EDMA were 1:1:4 and 70%, respectively. Its AC and TE factor were 8.37 and 7.68, respectively. Equilibrium dissociation constant at higher affinity site and lower affinity site were KD1=120.63μmol L-1 and KD2=3.31μmol L-1, respectively. Maximum apparent adsorption capacity at higher affinity site and lower affinity site were Qmax1= 141.13μmol g-1 and Qmax2=19.97μmol g-1.In the third part, a high-performance liquid chromatography (HPLC)-ultraviolet detection method, combined with MIP-solid-phase extraction (MIPSPE), was developed for the determination of nineβ-diketone antibiotics including PIP, FLR, NOR, OFL, TC, OTC, CIP, ENR, LOM in surface and waste water samples. Extraction of the nine components was carried out with the MIP cartridge. The water sample was added in Na2EDTA and adjusted to pH=6.5. Then the cartridge was eluted by a mixture of methanol and acetic acid (3mL,6:1/v:v). Separation of the nine components was achieved by using an Agilent HPLC and detected by a UV detector at a wavelength of 275 nm. Recoveries of the nine components in the spiked sewage sample were between 75.2 and 97.9%with a relative standard deviation of less than 7.1%. The limits of detection (S/N=3) were between 1.2×10-8~1.0×10-7 mol L-1. The proposed method was applied to determine the waste water of Sibao sewage treatment plant (STP). Results indicated that four components were detected in influent of Sibao STP, the concentrations were between 3.4~67.8×10-10 mol L-1. While in effluent of Sibao STP there were three components were detected, the concentrations were between 5.0~47.2×10-10 mol L-1.
