Abstract

The use of lactic acid bacteria fermented food has a long history, by lactic acid fermentation processed foods can not only improve food flavor, improve the nutritional value, but also has special nutritional value and physiological activity. A large number of studies have shown that lactic acid bacteria can regulate the body's normal flora of the gastrointestinal tract, thus inhibiting the growth of spoilage bacteria in the gut and corruption product generation; produce bacteriocins and other metabolites also inhibit bacteria growth and other effects. Because of its efficient, non-toxic, no residue, no drug resistance and other advantages, to feed to improve meat quality, feed conversion rate. Screening and isolated a strain of lactic acid bacteria in our laboratory, and its physiological and biochemical characteristics of studies have shown that it is most similar to L. plantarum (Lactobacillus plantarum), we will be named Lactobacillus plantarum ZJ316. Produce bacteriocin named Plantaricin ZJ316. Primarily for the separation and purification of bacteriocins and application of a preliminary study, include: determination of the ZJ316 antibacterial spectrum of Lactobacillus plantarum. The experimental results show that the strain has a broad-spectrum antimicrobial activity, after the double-layer tablet antagonistic Determination of the law, found that it also has the ability of anti-fungal. Turn Lactobacillus plantarum ZJ316 separation and purification of bacteriocins. First, aqueous two-phase extraction method for the initial separation of PEG / ammonium sulfate system optimum extraction conditions the the PEG4000 concentration is 17.5%, 19% (NH4) 2SO4 concentration, sodium chloride concentration of 1%, pH value of 3.75, The experiment proved that the phase extraction under this condition titer: 287.56IU/mL. PEG / sodium sulfate aqueous two-phase extraction system in the optimal experimental conditions for bacteriocin is the PEG molecular weight is 1000, and the concentration of 17%, the sodium sulfate concentration of 15%, the concentration of sodium chloride is 1.0%, pH 3.5, and tested Validation Under this condition, the extraction phase titer: 256 IU / mL. Selected ion column purification. Tested to confirm, select a pH of 5.0, sodium acetate buffer, the DEAE Sepharose FF packing purified and isolated finally obtained a relatively pure bacteriocin component. After using Trcine-SDS-PAGE electrophoresis detection, according to preliminary estimates electrophoresis results plantaricin ZJ316 molecular weight of about 6.5 kDa, belongs to a small molecule peptide substance. Preliminary exploration Lactobacillus plantarum ZJ316 as a feed additive used in piglet feeding trial. The results show that: add Lactobacillus ZJ316 fed piglet daily gain, feed conversion rate was more visible; diarrhea rate and reduce mortality, can play the antibacterial effects of alternative antibiotics; pork quality improvement role. Paraffin sections of the small intestine villi morphological changes observed, we can see that the piglet small intestine of Lactobacillus plantarum ZJ316 certain, from a physiological point of view supported by the experimental group piglet, feed conversion and meat quality were better than the control group conclusion.
