This paper studies by recombinant Escherichia coli BL 21 fermentative production of human-like collagen protein phase Ⅱ expression of pyruvate added to improve the human-like collagen Ⅱ production. Response surface methodology and the use of the process of adding the effect of pyruvate class Ⅱ collagen production of human factors ( added amount of pyruvate , pyruvate phosphate buffer, adding time and concentration) for the shake flask experiments optimized. Finally, in a phosphate buffer concentration is 0.17mol / L of fermentation medium , adding the induction 8.23mmol / L pyruvate human collagen Ⅱ was obtained The yield was 0.29g / L, the experimental group compared to the control growth 11.54% , the cell OD60 ¨ 0 than the control group increased 28.72% . In shake flask on the basis of experimental data , with the bioreactor added amount of pyruvate , pyruvate Added two factors on human collagen type Ⅱ in yield were studied . The results showed that : can significantly improve the human-like collagen Ⅱ production of pyruvic acid dosage 15.84mmol / L, pyruvic acid were added to the induction of one hour . In this condition, the cell dry weight and protein production reached 69.6g / L and 11.87g / L.
