Abstract


Ascorbic acid (L-ascorbic acid, AsA) that vitamin C is a small molecule antioxidants commonly found in plant tissue in the organism has important metabolic function and antioxidant effects. AsA human essential vitamins must be obtained from food because the body lacks the ability of synthetic AsA, and plants provide mankind with a major source of ascorbic acid. Ascorbic acid and its metabolic enzymes able to clear environmental stress generated by reactive oxygen species, which play an important role in plant resistance to environmental stress. Ascorbic acid biosynthesis and metabolism-related enzyme gene expression by regulating plants, you can improve the content of ascorbic acid, thereby increasing the nutritional value of plant products, and enhance the resistance of plants to Stress. Dehydroascorbate reductase (DHAR) plant ascorbic acid - a key enzyme in the glutathione cycle, catalytic reduction state of glutathione restore dehydroascorbic acid (DHA), DHA restore to restore the state of ascorbic acid. In this study, potato DHAR of two isoenzymes gene DHAR1 DHAR2 import the tomato it overexpression, to study the function of stress in tomato of AsA metabolism and adversity. The main results are as follows: 1. Potato DHAR1 and justice DHAR2 expression vector, Agrobacterium-mediated transformation of tomato. With kanamycin resistance to transgenic plants detected by PCR and fluorescence quantitative PCR results prove successful the tomato plants to turn of justice DHAR1 and DHAR2 gene. DHAR activity DHAR1 overexpression transgenic plants and fruit, AsA content and AsA / DHA than the wild-type increased significantly. DHAR2 super expression in transgenic plants leaves of DHAR activity, AsA content and of AsA / DHA significantly increased fruit, these indicators did not change that DHAR2 only organs containing chloroplasts of expression, its leaves super expression did not affect fruit DHAR activity and AsA content. From 3. DHAR1 and DHAR2 the overexpression could improve tomato leaf chlorophyll content and photosynthetic rate. Description DHAR overexpression important role in the stability of chlorophyll, enhanced photosynthesis. 4 overexpression DHAR1, DHAR2 transgenic plants for resistance to salt stress and methyl viologen stress improved to some extent. Under salt stress, overexpression DHAR1, DHAR2 transgenic plants compared with wild-type plants, have higher germination rate, plant height, stem length and chlorophyll content. High concentrations of methyl viologen oxidative stress, the transgenic plants leaf discs necrosis less than wild-type plants, chlorophyll content than the wild-type higher. Under the conditions of low temperature treatment, compared with the wild-type plants, overexpression DHAR1 gene in tomato leaves DHAR activity of AsA and chlorophyll content increased cell membrane permeability, membrane lipid peroxidation product MDA content reduced. Overexpression DHAR1 transgenic plants to cold resistance have improved to some extent.
