Abstract


Wheat (Triticum aestivu) is an important food crop. In order to adapt to a variety of abiotic and biotic stress on wheat yield, a wide variety of new varieties came into being. One way to increase the genetic variability of the wheat is the somatic cell hybridization. It can be from other genera or species the introduction of new features. Aegilops in wheat distant family plants, it may be a useful trait for wheat, especially in the resistant attention of agronomy characteristics. Intergeneric hybridization incompatible, the only way to get these intergeneric hybrids protoplast fusion and cell hybridization. Immature embryos of wheat and goat grass leaves, the test for the study material, the choice of the three dyes: with neutral red, FDA and rhodamine B, respectively, of its enzymatic protoplast stained, and 1 × 106 were observed, 5 × 106,1 × 107,5 × 107 protoplasts / mL four density stained fusion rate, and ultimately to determine the best combination of processing. CPPU has been widely used in fruit trees, melons, vegetable production, but few studies in wheat production. 5-azaC and CPPU on different periods of wheat by seed treatment and foliar spraying of way, and take advantage of the high performance liquid chromatography and other analytical tools, a comprehensive study of the different treatments on aging indicators, wheat production and from the point of view of DNA methylation, further 5-azaC and CPPU improve the wheat production mechanism study, the main findings are as follows: wheat seeds by the low concentration (5-500μM) 5-azaC treatment reduced DNA methylation levels could induce paksaylabgg, Taichung29 and Taichung29 * 6/CarstensV, three kinds of wheat flowering. 1000μM 5-azaC treatment of wheat seedlings died after 20 d. Flowering stage foliar application of 5-azaC, anti-aging, reduce the level of DNA methylation of the flag leaf, the contribution rate of the stems and leaves of grain and a slight increase in the protein content and gluten content in wheat grain . CPPU can delay the aging of the flag leaf, reduce the level of DNA methylation of the flag leaf and single stem panicle weight, promote transit of organic material to the library, so that the increase in protein content and gluten content in wheat grain.
