Abstract


Cryopreservation of plant germplasm resources and long-term preservation of the most effective methods . In this paper, to spearmint (Mentha spicata L.) shoot tip material, its vitrification cryopreservation conditions , another study the physiological and biochemical reactions Cryopreservation of spearmint material , genome stability and DNA methylation role. The main findings are as follows : 1 spearmint shoot tips by vitrification cryopreservation program of exploration and research results show that : the the 4 ℃ exercise 3-4 weeks , add 2 mol · L -1 < / sup pre - cultured for 3 days in > glycerol MS medium the liquid pretreatment culture at room temperature for 20 min , PVS2 0 ° C dehydration of 50 min , the highest survival rate , up to about 60% . Normal regenerated plants differentiation . 2 spearmint by ultra-low temperature processing malondialdehyde (MDA) were significantly elevated , peroxidase (POD), catalase (CAT) activity were significantly enhanced compared with the control group . The physiological and biochemical indicators illustrate , ultra-low temperature processing affect spearmint normal physiological and biochemical reactions leading to the accumulation of malondialdehyde (MDA) , some changes in the activity of antioxidant enzymes . 3 use the amplified fragment length polymorphism (AFLP) technique for processing the recycled material and the genomic DNA of the control group were analyzed, between the treatment group and the control group was not found different fragments . Did not cause changes in the save material genomic DNA sequence showed that in the cryopreservation process . Methylation levels between 4 methylation - sensitive amplification polymorphism (MSAP) marker technology analysis and processing with the control group sample . MSAP technology methylation of ultra-low temperature processing of the material before and after analysis showed that : compared with the control , ultra-low temperature processing of recycled materials methylation status and changes in the level of the lower level of genome-wide DNA methylation of cytosine , another methylation and demethylation of 8.93% and 12.3% , respectively .
