Abstract


Objective: To investigate recombinant human erythropoietin in neuronal degeneration, cell proliferation and mossy fiber sprouting in the adult rat hippocampal dentate of status epilepticus back. Methods: (1) modeling and grouping: selection of 140 clean healthy adult male SD rats were randomly divided into normal control group of 20 sterile saline, intraperitoneal injection of the same amount, more than 120 lithium chloride - cefpirome card products epileptogenic test production model of temporal lobe epilepsy, and in accordance with the the Racine standards determine the extent of seizures were graded, select epileptic state (SE, statusepilepticus) continued 1h and above selected, and then randomly divided into recombinant human erythropoietin erythropoietin (Recombinant Human Erythropietin, r-HuEPO) SE group (intervention group) and SE (not intervention group), the intervention group in SE 6h after each re intraperitoneal injection of r-HuEPO 5000IU /, will ultimately survival model The rats used in the next test, each each experimental animals are four, as the study site in the hippocampal dentate gyrus; (2) four days after modeling FJ-B (Fluoro-Jade B) line immunofluorescence detection and observation of the dentate gyrus of the hippocampal neuronal degeneration; (3) drawn in 9 days and BrdU (5-bromodeoxyuridine, 5 - bromo-deoxy-uridine) immunohistochemical detection observed dentate back cell proliferation; Timm's stain drawn in 14 days to observe the dentate gyrus mossy fiber sprouting; drawn BrdU immunohistochemistry and Timm's stain (4) 28 days. Results: (1) the experimental rats by intraperitoneal injection of lithium chloride - pilocarpine, SE induced a success rate of 88.57%, 70.17% of the total mortality; (2) FJ.-B staining showed: SE fourth day hippocampal dentate gyrus degeneration of nerve elements in the SE group than in the SE r-HuEPO group and the control group significantly, especially SE group, mainly in the dentate gyrus SGZ (subgranular zone, granulosa cell area) and gate area: SE group ( P lt; 0.01, SE group with SE r-HuEPO group (11.12 ± 0.75) 32.32 ± 5.09) compared with the control group (6.50 ± 1.08) compared to P lt; 0.01, SE r-HuEPO group and the control group than the P gt ; 0.05; (3) of BrdU immunohistochemical staining results: SE 9th day of immunization of BrdU-positive cells in sE group increased significantly, mainly focus on particle zone, was the cluster distribution; significant increase SE r-HuEPO group still mainly distributed particles under the district, were scattered; control group also have very small amount of immune reaction positive cells scattered; SE group (92.8 ± 2.97) and the control group (12.80 ± 2.13) than the P lt; 0.01, SE group and SE r-HuEP0 (16.95 ± 0.54) groups than P lt; 0.01, SE r-HuEPO group and the control group than in the P gt; 0.05; 28 days, BrdU immunoreactive cells further increased significantly in the SE group, and a lot to the door migration zone, part of the distribution to the granule cell layer and inner molecular layer, BrdU immunoreactive SE r-HuEPO group, although the number of positive cells increased and also part of the migration to the door area, but no significant increase; control group BrdU immunoreactive cells increase not obvious, but still the main zone of the particles, the SE group (146.45 ± 5.91) and control group (38.52 ± 3.10) than P lt; 0.01, SE Group SE r-HuEPO group (46.27 ± 2.93) than P lt; 0.01 SE r-HuEPO group and the control group than the P gt; 0.05; (4) Timm's staining results: SE after 14 days, each group Timm's stain particles increase is not obvious, but the SE group (0.45 ± 0.19) and the control group (0.10 ± 0.11) than P lt; 0.05; the SE group mossy fiber germination obvious in the first 28 days, and the inner molecular layer connection into a dense strip SE r-HuEPO group score has also increased, and the control group was not significantly different ; the SE group (3.60 ± 0.16) and the control group (0.30 ± 0.11) than P lt; 0.01, SE and SE r-HuEPO group (0.55 ± 0.10) than P lt; 0.01, SE r-HuEPO group and the control group than the P gt; 0.05. Conclusion: (1) status epilepticus induced rat dentate gyrus neuronal degeneration and cell proliferation, and the large number of newborn cells migrate to the gate area, the migration of some inward molecular layer. Will also result in the mossy fiber sprouting in the dentate gyrus neurons, the molecular layer of the the germination fiber inward extension. (2) human erythropoietin male SD rats after status epilepticus seizures 7-8W age, to reduce the mortality rate is not statistically; (3) erythropoietin status epilepticus seizures of large mouse, can reduce the proliferation of neuronal degeneration and status epilepticus seizures, and can inhibit mossy fiber sprouting in the dentate gyrus.
