Objective: Epilepsy is a common chronic disease of the nervous system, genetic factors is one of the causes. Human genetics studies often compare crowd if it contains a particular trait genes lead to identify and analyze the reasons for the differences between the different groups of people. Genetic research can eventually discovered the the responsibility gene (or genome), especially when unable to obtain biological information to explain the origin of the disease, the success of genetic research is the idea of ??widening the researchers found completely unknown in the past or not. suspected of being associated with the disease process possible. The purpose of this experimental study is a 5 - HT 2A receptor (5-HTR2A) gene expression in idiopathic epilepsy patients with lesions in the frontal and temporal lobe brain tissue with the control group, the frontal and temporal lobe, and explore with epilepsy relationship of pathogenesis, clinical diagnosis and treatment, prevention provide a theoretical basis. Method: the experimental group specimens from Chengdu sixty-three hospitals neurosurgery epileptic focus resection patients before surgery by the long-range video EEG monitoring, the dipole position clear epileptic focus in the frontal lobe and temporal lobe endorsement for patients and their families have been informed book . Etiological classification in accordance with epilepsy, all patients with idiopathic epilepsy, no significant epileptogenic etiology, no focal neurological examination positive signs, radiographic examination no positive findings, more than 2 years duration, after poor efficacy of multidrug therapy , and in accordance with the diagnostic criteria of refractory epilepsy. Which the frontal lobe of 13 cases, with an average age of 21.77 years old; temporal lobe in 22 cases, with an average age of 24.75 years old. The control group specimens were from six to three hospitals neurosurgical intracranial decompression patients. Frontal lobe brain tissue in 11 cases, with an average age of 51.45 years; temporal lobe brain tissue of three cases, with an average age of 27.33 years old. The experiments using real-time quantitative PCR dye assay 5-HTR2A expression in the frontal and temporal lobe brain tissue of the experimental group lesions prefrontal and temporal lobe brain tissue and control group, the difference of the two groups were compared using independent samples t-test between sex. Results: (1) 5-HTR2A average in the frontal lobe of the epilepsy group and the control group the relative expression intensity were 8.72 ± 3.75 and 3.71 ± 2.14, by t-test (t = 2.95) was probability P value of 0.009, the amount of the epilepsy group 5-HTR2A expression lobe brain tissue is 2.35 times that of the control group, the results were statistically significant (P lt; 0.05). (2) 5-HTR2A average in the epilepsy group and the control group temporal lobe brain tissue relative expression intensity were 0.132 ± 0.073 and 0.121 ± 0.024 by t-test (t = 0.143) was the probability P value of 0.888 (P gt; 0.05) between patients with epilepsy and control group, indicating 5HTR2A in little change in the expression level of the temporal lobe, between no significant difference (P gt; 0.05). Conclusions: (1) the physiological state 5-HTR2A expression level of the frontal lobe in the brain larger than the temporal lobe. (2) 5-HTR2A gene expression in the frontal brain tissue of patients with refractory epilepsy was significantly higher than that of non-epileptic patients, the 5-HTR2A gene upregulation may be related to the occurrence of epilepsy, especially the occurrence of refractory epilepsy There are close relations. Might have speculated that the 5-HT2A receptor in the central role of enhanced cell excitability, increased expression of the receptor may be one of the causative factors of epilepsy.
