Diabetes is a group of metabolic diseases characterized by increased blood glucose levels in chronic group, is a common disease. According to WHO statistics, the end of the 1970s, the early 1980s, the global diabetes of more than 30 million patients with diabetes has reached 150 million by the mid-1990s, the global figure will double again by 2025. Our survey shows that diabetes patients: from 1979 to 1980, China's adult diabetes prevalence rate of 1% from 1994 to 1995, the prevalence rate of 2.5%. Estimated that about 30 million of our existing patients, ranking second in the world. In recent years, with the improvement of people's living standards, population aging, lifestyle changes, the incidence of diabetes is increasing rapidly. Diabetes Candida vaginitis diabetic patients in several common vaginitis one of the highest incidence of diabetic women most likely to suffer Candida vaginitis. Diabetes Candida vaginitis and difficult to cure, easy recurrence. Thus, in the context of the prevalence of diabetes increased year by year, to investigate the pathogenesis of diabetes Candida vaginitis, looking for ways to effective prevention and treatment of diabetes Candida vaginitis is important to improve the quality of life of women. The study found that estrogen induces Candida albicans vaginitis in a mouse model, TGF-β1 was expressed at high levels, while TGF-β1 is an immunomodulatory factors inhibit the cell-mediated immune response. The study found that TGF-β1 plays an important role in the development of diabetes and its many complications. Thus, we speculate: TGF-β1 may play a role in the pathogenesis of diabetes Candida vaginitis. Therefore, the purpose of this experiment is to study whether TGF-β1 in diabetes Candida vaginitis occurrence and development work, in order to explore the possible pathogenesis of diabetes Candida vaginitis. The first part of the diabetes and diabetes Candida vaginitis rat model building Objective: To establish a rat model of diabetes and diabetes Candida yeast vaginitis rat model, in preparation for the follow-up observation of the expression of TGF-β1. Methods: 26 female SD rats (weighing about 200g) fasted for 12h after intraperitoneal injection of streptozotocin (STZ) 65mg/kg. Candida albicans inoculated one week after the the the rat vaginal lavage visible spores and moderate amount pseudohyphal fungal culture results showing that a large number of white colonies growing. Conclusion: The experiment successfully established diabetic rat model of diabetes Candida yeast vaginitis rat model. Purpose of the expression of TGF-β1 in the second part of the diabetes and diabetes Candida vaginitis rat model of vaginal tissue: detection of diabetes and diabetes Candida yeast vaginitis vaginal tissue in rats, the level of expression of TGF-β1 to explore the TGF -β1 induced diabetic Candida vaginitis possible mechanisms. Methods: 36 female SD rats (weighing about 200g) were randomly divided into diabetic group (D group, n = 26) and the control group (C group, n = 10). Diabetic rats by intraperitoneal injection of streptozotocin (STZ) 65mg/kg. Blank control group, a single intraperitoneal injection of the same amount of citric acid - sodium citrate buffer solution. A week later, into diabetic rats and mold blank control group rats were randomly selected five killed take the vaginal tissue. Remaining molded diabetic rats were randomly divided into diabetes Candida vaginitis group (DC group, n = 10) and indomethacin treatment of diabetes Candida vaginitis group (DI group, n = 10). The remaining blank control rats as a non-diabetic patients with Candida yeast infection group (CC group, n = 5). The three groups were inoculated with C. albicans yeast spores 5 × 10 8. The DI group on vaccination day after vaginal indomethacin treatment for 6 days (2m M, 100ul) and the other two groups were given the same amount of sterile saline. One week after vaccination, the rats were killed, and the vaginal tissue after vaginal lavage. Lavage for microscopic examination and fungal culture. ELISA was used to detect the levels of TGF-β1 in rat vaginal tissue. Results: DC visible to a large number of spores and a moderate amount of pseudohyphae; DI were observed in a small number of spores but no pseudohyphae; CC group no spores. DC visible to a large number of C. albicans colonies growing; DI group showed a small amount of Candida albicans colony growth; CC group showed colony growth. The D rats vaginal tissue TGF-β1 levels were significantly higher than the control group, P lt; 0.05, the difference was statistically significant. DC rats CC group was significantly higher than the level of TGF-β1 in the vaginal tissue, P lt; 0.05, the difference was statistically significant. DI rats compared with the DC group level of TGF-β1 in the vaginal tissue, P gt; 0.05, the difference was not statistically significant. Conclusion: TGF-β1 induced diabetes Candida plays an important role in the occurrence and development of vaginitis.
