Effect of Global Cerebral Ischemia-reperfusion to Tight Junction Protein Occludin in Rat Brain

Abstract


Objective four artery occlusion established global cerebral ischemia -reperfusion model , rats reperfusion injury tight junction protein occludin expression level of the brain tissue . 30 healthy male SD rats were randomly divided into sham operation group , ischemia 30 minutes reperfusion 3h , 12h , 24h , 72h , a total of five groups of six each . Sham group only take bilateral vertebral artery coagulation , ischemia-reperfusion group electrocoagulation bilateral vertebral artery and occlusion of bilateral common carotid artery for 30 minutes , then open bilateral carotid artery and ischemic perfusion point in time to kill rats and the brain , Western Blot and RT-PCR reaction occludin changes from the protein level and gene level . Results ischemia reperfusion group compared with the sham group of Occludin protein and mRNA levels decreased (P lt; 0.05), reperfusion 3h began to decline , 12h decreased the minimum level , when reduced to 24h , 72h appear slow recovery , but still below the level of the sham group . The comparison was statistically significant ( P lt; 0.05 ) in the experimental group group . Conclusion rat brain ischemia-reperfusion tight junction protein occludin expression levels decreased , and with changes in protein and mRNA levels dropped by an average of reperfusion time .
