Abstract


Objective: To investigate the expression of P-aPKC-ιand E-cadherin in cholangiocarcinoma, compare to its adjacent tissues and congenital choledochal cyst, and analyze the role of P-aPKC-ιand E-cadherin in the tumorigenesis and development of cholangiocarcinoma. Provide a preliminary basis for further study of the mechanism of aPKC-ιin the cholangiocarcinoma.Methods: Collect the surgical removal and pathologically confirmed cases of cholangiocarcinoma, record the hospitalization number, pathology number, sex, age, pathological results, differentiation, TNM stage, and gather benign lesions choledochal cyst as control. Access to 37 cases of cholangiocarcinoma and 10 cases of congenital choledochal cyst with complete data. The expression of P-aPKC-ιand E-cadherin were detected by SABC immunohistochemistry in cholangiocarcinoma tissue, cancer tissue (from cancer> 2cm) and congenital choledochal cyst tissue. Read the Slices in semi-quantitative way and use SPSS13.0 software for statistical analysis.Results:1, P-aPKC-ιwas low or even no expressed in biliary cyst tissue (8/10, 80%) and adjacent tissue of cancer(29/37, 78.38%), highly in bile duct carcinoma (24/37, 64.86%), sited in the cytoplasm and nucleus. The expression of P-aPKC-ιbetween choledochal cyst and adjacent tissue was no significant difference (P=1.0000), but compared with cholangiocarcinoma, was statistically significant (P= 0.030, P<0.001). The expression of P-aPKC-ιand TNM stage and differentiation of bile duct cancer was positively correlated (P = 0.002, P = 0.001).2, E-cadherin was highly expressed in the common bile duct cyst (9/10, 90%) and adjacent tissues of cancer (27/37, 72.97%), mainly location in the apical membrane side, low or loss expressed in the bile duct carcinoma (25/37, 67.57%). The expression of E-cadherin between choledochal cyst and adjacent tissue of cancer was no difference (P=0.479), while the expression of them compared with cholangiocarcinoma, the difference was statistically significant (P=0.004, P<0.001). The expression of E-cadherin and TNM staging and differentiation of cholangiocarcinoma was negatively correlated (P=0.035, P=0.017).3, The expression of P-aPKC-ιand E-cadherin was associated, the expression of the P-aPKC-ιwas low while E-cadher was high in the common bile duct cyst; the expression of P-aPKC-ιwas high but E-cadherin was low or loss in the cholangiocarcinoma. All of the above prompted that the two may play a role in the tumorigenesis and development of cholangiocarcinoma together.Conclusion:1, The expression of P-aPKC-ιwas high in cholangiocarcinoma tissues, while low in non-cancerous tissues; the expression of E-cadherin was high in non-cancerous tissues, but low in cholangiocarcinoma tissues. The two suggested that activated aPKC-ιwas associated with the tumorigenesis of cholangiocarcinoma, and may play a synergistic role with E-cadherin in this process.2, High expression of P-aPKC-ιand low of E-cadherin were associated with The TNM stage and pathological differentiation of cholangiocarcinoma, suggesting that activated aPKC-ιtogether with E-cadherin may also play a role in the development of cholangiocarcinoma.
