Abstract


ObjectiveExpression of ERCC1 (Excision Repair Cross Complementing 1) protein in the specimens of chronic hepatitis b (CHB) and cirrhosis after HBV infection were both detected with immunohistochemistry (IHC). The relationship between the expression status of ERCC1 and the clinical pathological characteristics was analyzed. And the differences between the expression of ERCC1 and different degrees of inflammation and liver fibrosis were compared in order to elucidate its function in the progression to the liver fibrosis. even to the hepatocellular carcinoma.MethodsAll the cases were devided into different groups according to different degrees of inflammation and liver fibrosis. ERCC1 expression status of 73 patients with CHB and 13 cases specimen of cirrhosis after HBV infection were detected by SupervisionTM two-step method of IHC. SPSS 18.0 statistical software was applied for data analysis, with P<0.05 as the level of significance. The correlation between the expression of ERCC1 and the major clinical pathological characteristics of specimen were analyzed by Chi-square test and Kruskal-Wallis H test. The relationship of two variables in the experiment was analyzed by Spearman test, with P<0.05 as the level of significance.Results1. ERCC1 protein expression was mainly observed in the liver cell nucleus of both liver specimens. The positive rate of ERCC1 protein expression in 86 cases of liver tissues was 32.6%. The positive rate of ERCC1 protein expression in CHB specimen was 31.5%, while its positive rate in cirrhosis’specimens was higher. The positive rates of ERCC1 protein expression between the two specimens were no significance (P>0.05).2. Comparison between different groups:(1) The expression of ERCC1 protein increased with inflammatory activity increasing at the beginning, but decreased later. The difference was not statistically significant (P>0.05). (2) The expression of ERCC1 protein increased with the level of liver fibrosis increasing at the beginning, but decreased later. The difference was not statistically-significant (P>0.05).3.The positive expression rate of ERCC1 in different gender, age, nation, first-degree family history of hepatitis, smoking history, alcohol history, ALT, AST, T.Bil, Alb, A/G, PTA, e antigen, e antibody, AFP status and HBV-DNA quantification, respectively, was not significantly difference (P>0.05, respectively). But the positive expression rate of ERCC1 in different HBV-DNA status was significantly difference (P<0.05), the positive expression rate of ERCC1 in group HBV-DNA (+) was lower than group HBV-DNA (-), and there was a negative correlation between them.Conclusion1. The positive expression rate of ERCC1 was not significantly correlated with degrees of inflammation or degrees of fibrosis. 2. The positive expression rate of ERCC1 in group HBV-DNA (+) was lower than group HBV-DNA (-), and there was a negative correlation between them (P<0.05). And the mechanism of interaction between them still need further study.3. The expression of ERCC1 was not significantly correlated with gender, age, nation, first-degree family history of hepatitis, smoking history, alcohol history, ALT, AST, T.Bil, Alb, A/G, PTA, e antigen, e antibody, AFP status and HBV-DNA quantification, respectively.
