Abstract


The purpose of the experiment through the establishment of two different periods of prenatal stress rat model, observed in different periods of prenatal stress on behavior of offspring rats learn, the cytokine interleukin-18 (interleukin 18, IL-18) and extracellular signal-regulated kinase (Mitogn activated protein kinase kinase 1, MEK-1) the impact, so as to explore the pathophysiological mechanisms of the effect of prenatal stress sensitive and gender, to prenatal stress in the pathogenesis of the disease basis. Methods SD pregnant rats were randomly divided into three groups, the control group during pregnancy does not give any stimulation; interim stress group given the shackles of stimulus from 9-14 days of pregnancy; late pregnancy group from 15-20 days of gestation bound to give stimulus . Until the offspring born 21 days after weaning, and male and female rats were housed separately. Adult offspring as soon as 75 days after birth were open chamber experiments, new things, experiment, the sucrose favorite experiments, prepulse inhibition and fear reflex test and forced swimming test five kinds of behavioral testing and detection of hippocampal IL-18 and MEK-1. Results a. Behavioral experimental results: 1. Openfield experiment: The results showed that, compared with the control group for male offspring stimulate male offspring in the third trimester prenatal openfield away to reduce (p lt; 0.05), the total distance of the central region to reduce (p lt; 0.05), the central region of the residence time of reduction (p lt; 0.01). 2. Something new experiment: the results show the time of exposure to new things for male offspring of the control group compared to the male offspring in the second trimester perinatal stress group for male offspring (p lt; 0.01) and third trimester perinatal stress group (p lt; 0.05) are reduced. 3 sugar consumption experiment: The results showed that sugar consumption to reduce the average weight of male offspring in the second trimester prenatal stress group (p lt; 0.05) compared with the control group for male offspring. 4 prepulse and panic inhibition experiments: The results showed that the first pulse suppression offspring of the reaction in the second trimester prenatal stress group compared with the control group offspring reaction reduce (p lt; 0.05); third trimester prenatal panic reaction should excited offspring response to increased offspring compared with the control group (p lt; 0.01). 5. Forced swimming test: The results showed that the third trimester prenatal stress group compared with the control group offspring female offspring (p lt; 0.05) and male offspring (p lt; 0.01) \second trimester prenatal stress group only male offspring rats \Dorsal hippocampus and ventral hippocampal IL-18 content and MEK-1 positive cells in test results: 1.IL-18 results showed that compared with the control group, the third trimester prenatal stress can lead to female offspring ventral hippocampus IL-18 was increased (p lt; 0.05), while the second trimester prenatal stress offspring hippocampal IL-18 content affect (p gt; 0.05). 2.MEK-1 positive cells results show that compared with the control group (p lt; 0.01) and ventral hippocampus of MEK-1 expression decreased (P LT; 0.05) third trimester of pregnancy can result in the dorsal hippocampus of MEK-1 expression increased. the second trimester of MEK-1 expression in the hippocampus affect (p gt; 0.05). Conclusion 1. Perinatal stress of the second trimester and third trimester of pregnancy can lead to changes in offspring behavior, but the third trimester prenatal stress on offspring behavior learned more. Prenatally stressed offspring rats of different sex behavior changes of the differences in the impact of prenatal stress on male offspring than female offspring. 3. Third trimester prenatal stress can lead to the offspring rats hippocampus inflammatory cytokines IL-18 secretion increased and MEK-1 signaling protein abnormal expression. Ventral and dorsal hippocampus \5.IL-18 may be involved in the regulation of the MEK-1 signaling protein expression.
