Abstract


Background: Diabetes is a chronic metabolic disorder that has now become a serious threat to human health, the third largest of chronic non-communicable diseases. In recent years, the incidence of diabetes is increasing every year, compared with non-diabetic population, the high prevalence of the diabetic population in the atherosclerotic disease, younger age at onset, rapid progression of multiple organ involvement at the same time more . In addition, atherosclerosis caused by cardiovascular and cerebrovascular diseases is one of the main reasons for death and disability in patients with diabetes. Numerous studies have confirmed the early stage of diabetes, presence of atherosclerosis, the two are closely related. Recent studies have found that chronic inflammatory reaction is one of the important factors to lead and promote diabetic atherosclerosis, TCM theory holds that phlegm, blood stasis is the main pathological product of low-grade inflammation, phlegm and low-grade inflammation underlying pathology characteristics of the diabetic Apo E gene knock in addition to the study mice coronary atherosclerosis inflammatory response to the heat and activating blood compatibility of traditional Chinese medicine to provide the scientific basis for new ideas in the treatment of diabetic coronary atherosclerosis. Objective: By observing the heat and activating blood compatibility of traditional Chinese medicine for diabetes Apo E gene knock on impact in addition to the mouse coronary atherosclerosis inflammatory response, to provide a scientific basis for clinical use in the treatment of diabetic coronary atherosclerosis with new ideas. Methods: Apo E gene knockout mice from 8 weeks of age to be a high-fat diet, and be streptozotocin intraperitoneal injection manufacturing diabetes model (40), after modeling heat and activating blood compatibility were randomly divided into treatment group (20 ) and the control group (20). Heat and activating blood compatibility of traditional Chinese medicine 0.9g/kg d and NS lOmg / kg · d orally from 13 weeks after administration were given 16 weeks after administration, blood glucose, blood cholesterol and serum glycosylation end product (AGEs) level assessment of coronary artery pathomorphology picrosirius staining methods coronary collagen content, HE staining coronary stenosis opening percentage luminal occlusion incidence and coronary surrounding inflammatory infiltration degree of immunohistochemical determination of plaque monocytes / macrophages -2 (MOMA-2) expression, ELISA method for determination of plasma ICAM-1 expression, Real-time PCR method for determination of ICAM-1, MCP-1 , HO-1 expression. Results of: heat blood circulation compatibility TCM group compared with the control group, coronary arteries stenosis rate (37.3% ± 3.87%, 43.1% ± 4.99%), coronary arteries occlusion rate (17.2% ± 1.00%, 29.3 ± 0.73% ), coronary surrounding inflammatory cell density (17.9% ± 1.97%, 26.1% ± 2.77%), MOMA-2 content (29.09% ± 3.12%, 58.06% ± 5.49%), coronary collagen content (5.23% ± 0.63% , 13.77% ± 1.05%) are significantly significant difference (P lt; 0.01); addition, clearing heat and activating blood compatibility of Chinese Medicines mice after 15 weeks of fasting blood glucose levels, serum advanced glycosylation end product level with the control group comparison also have decreased significantly ( P lt; 0.05); of heat blood circulation Combined with Herbal also significantly reduce ICAM-1, MCP-1 gene expression (P lt; 0.05), and significant expression increased HO-1mRNA. (P lt; 0.01). Conclusion: heat and activating blood compatibility of traditional Chinese medicine can be a significant inhibition the diabetic atherosclerosis within the coronary arteries in mice, the outer membrane of the inflammatory response and the expression of inflammatory cytokines, lower main coronary artery stenosis and occlusion rate, slowing coronary atherosclerosis progress.
