Objective: a rat model of chronic stress Liver Qi Stagnation observed liver qi stagnation model behavior in rats, functional brain imaging, hypothalamic - pituitary - adrenal (HPA) axis hormones change and electricity for its intervention the impact, focusing on the use of positron emission tomography (PET) technology, objective visibility based on changes in brain function due to liver syndrome, and reveal the electro-acupuncture central mechanism of regulation of the certificate of impaired brain function. Methods: chronic stress combined with established rat model of liver syndrome solitary raised a total of four weeks. 2 according to the weight of the rats were randomly divided into three groups: normal group, model group and EA group (n = 10). Open field test (Open-Field Test), sucrose water partial addicted experiment, body weight, food intake was measured and general mental state, fur, intellectual, locomotor activity and stool consistency such as observation, judgment liver syndrome model rats behavioral changes as well as the behavior of electricity for its intervention school. The 4 use E-plus animal PET scanning instrument uses a pair of fluorine-18 deoxyglucose (18F-FDG) as a tracer model group, the EA rats PET functional brain imaging. Image analysis and statistical functional brain imaging field of gold standard SPM software model group (pre-treatment) and model group (after processing) data, electro-acupuncture group (pre-treatment) and EA group (treatment) data, model group (treated) and EA group (treatment) data for the two-sample t-test (P lt; 0.001), respectively, corresponding FDG metabolic differences in brain regions, namely the brain activation maps to obtain Liver Qi Stagnation by induced changes in brain function objective visibility based on the central mechanism of regulation of the certificate of impaired brain function and reveal the electro-acupuncture. 5 use was determined by ELISA in serum corticotropin-releasing hormone (CRH), corticosterone (CORT) content, and the impact of the state judge Liver Qi Stagnation HPA axis and the EA interventions HPA axis. Results: 1 behavioral research results: after 4 weeks of chronic stress procedure, compared with the normal group, the model rats horizontal movement score and the vertical movement scores were significantly reduced (P lt; 0.01). Model group of sucrose water consumption sucrose water partial addicted degree significantly reduced (P lt; 0.01), slow weight gain, food intake was significantly reduced (P lt; 0.01). The above indicators were improved by electro-acupuncture treatment (P lt; 0.05 or P lt; 0.01). 2 PET findings: ① model group (before treatment) and model group (after treatment) related brain regions compared with pre-treatment, 28d modeling after abnormal glucose metabolism in rats of the model group change involves brain areas more SPM2 analysis: . Glucose metabolism reduce brain regions (P lt; 0.001): the left cerebral hemisphere: central gyrus central gyrus: the right hemisphere: anterior cingulate: bilateral hemisphere: posterior cingulate; left diencephalon : dorsomedial nucleus lateral nucleus. Increased glucose metabolism in the brain regions (P lt; 0.001): the left cerebral hemisphere: parahippocampal gyrus, middle temporal gyrus, superior frontal gyrus, the island long back: right hemisphere brain: the right side of the striatum (gray matter); : ventromedial nucleus ventrolateral nucleus; left cerebellum. (2) the model group (treatment) and EA group (treatment) related brain areas SPM2 analysis: electro-acupuncture intervention compared with the model group, the EA rats reduced glucose metabolism brain regions (P lt; 0.001): The left cerebral hemisphere: frontal gyrus, inferior frontal gyrus, superior parietal lobule, back to the central beside lobular corpus callosum; the left diencephalon: undetermined vaginal discharge nuclear lateral nucleus. Increased glucose metabolism in the brain regions (P lt; 0.001): the left cerebral hemisphere: the central gyrus, postcentral gyrus, middle temporal gyrus, orbital back to the right hemisphere of the striatum (gray matter);: frontal gyrus, medial frontal gyrus ; the right side of the diencephalon: the subthalamic nucleus. (3) EA group (pre-treatment) and EA group (treatment)-related brain regions SPM2 analysis: 28d processing after the EA group rat cerebral glucose metabolism within set threshold (P lt; 0.001) and treatment Compared with no statistically significant difference. 3 HPA axis hormones findings: 28d end of the experiment, the model rats serum CRH and CORT content with the normal group, there were significantly higher (P lt; 0.01); intervention of the electro-acupuncture, electro-acupuncture group CRH, CORT decreased compared with the model group, a highly significant difference (P lt; 0.01). Conclusion: Chronic Stress liver qi stagnation rat model with anorgasmia inquiry activity decreased, slow weight gain, reduced food intake, stool histological changes and abnormal behavior, in line with Liver Qi Stagnation characteristics of clinical symptoms, the EA Ganshu, on the door on the Liver Qi Stagnation symptoms has a significant improvement. 2 Liver Qi Stagnation in the relevant brain regions abnormal glucose metabolism, and abnormal glucose metabolism is closely related to its function, the objective visibility based on changes in brain function due to liver syndrome. EA Ganshu on the door can be significantly improved liver qi stagnation caused by abnormal glucose metabolism related brain areas. Overall performance has increased the role of to reduce brain regions glucose metabolism, brain regions with lower elevated glucose metabolism, prompted EA Ganshu, on the door of the bi-directional regulation of glucose metabolism of the brain regions is its treatment of liver qi stagnation the certificate of one of the central mechanism of liver syndrome HPA axis performance hyperactive state, the EA Ganshu on the door inhibit CRH, CORT excessive secretion, suggesting a benign regulatory role for the HPA axis, which is the electroacupuncture treatment the certificate role of mechanisms
