Abstract


Proteasome degradation pathway is an important cellular protein degradation pathways , proteasome abound in both the cytoplasm and the cytoplasm . Proteasome cells play an important role , it is involved in the cell cycle , apoptosis , signal transduction, transcriptional regulation of many physiological processes . The proteasomal is from the core part of the 20s proteasome activating factor constitutes the activation factor contain 19s, 1ls , and PA200 . REGγ as 11s a family , it can activate the basic active sites of the 20S proteasome . The traditional view REGγ protein activation 20s proteasome can degrade some peptides only , therefore , relative to other proteasome -activating factor , about REGγ research is relatively less . This is the first demonstration REGγ protein degradation of intracellular protein in 2006 XiaotaoLi et al study found REGγ protein can activate the the 20s proteasome degradation of the protein SRC - 3 . Since then they have studies to prove REGγ protein can drop the protein of p21, p19 , MDM2 , HCV, . These research results show that the the , REGγ involved in a large number of physiological processes in the cells . REGγ involved in various physiological processes, wherein the part of the principle is achieved through the regulation of the level of protein from the target protein . One of the important direction of the the REGγ target protein is also REGγ protein research . In this issue we explore various HPLC purification methods including: ion exchange, molecular sieve, affinity chromatography , hydrophobic interaction chromatography purification with high purity the 20s proteasome and REGγ of protein . Determine the purification the 20s proteasome and REGγ of protein for the polymerization of the state , in vitro will 20s proteasome and REGγ of proteins combine to form a degradation system , has demonstrated the use of the target protein p21 verify their activity ; while for the first time the use of in vitro experiments can prove REGγ protein degradation of viral proteins HCV . The degradation of these proteins proved that the activity of the degradation system . The establishment of this system provides the basis for the screening of the target protein , but also can be used for the study of protein modifications .
