Abstract


Tomato yellow leaf curl disease (Tomato yellow leaf curl virus, TYLCV) is popular worldwide, a devastating viral disease, has become the limiting factor in tomato production in the world. Tomato yellow leaf curl disease in recent years in our country have increased year by year, from south to north, rapidly spreading trend has caused a very serious loss of Chinese tomato farming. Breeding for disease resistance has been considered to be the most effective, the most direct way to solve the disease problem. Therefore, the test to 10 kinds of tomato yellow leaf curl disease resistant materials and two kinds of susceptible materials for the test material, identification of Tomato yellow leaf curl virus; paraffin sections and scanning electron microscope studies of tomato yellow leaf curl virus material resistance gene and the correlation structure of the tomato leaves; molecular identification to verify the identification results and field natural incidence of tomato yellow leaf curl disease resistance breeding work provides a theoretical foundation. The content and the results are as follows: 1 DNA sequence comparison in GenBank tomato yellow leaf curl virus have been reported, and the DNASTAR software design a pair of primers based on conserved sequence in the tomato plants suspected of being infected tomato yellow leaf curl virus PCR amplified a 800bp specific band clones were sequenced and compared in GenBank, the establishment of the tomato yellow leaf curl virus identification method, and confirmed the infection experiment with the the tomato material of virus to tomato yellow leaf curl virus. 2 according to the growth of the tomato, and tomato yellow leaf curl disease prevalence, take leaves for histological examination and scanning electron microscopy. The test results show that: the PCR method can be quickly and accurately identified Tomato yellow leaf curl disease resistance, and homozygous Ty-1 and Ty-3 resistance gene in tomato material containing both higher and more stable resistance; tomato yellow The leaf curl disease, Ty-1 and Ty-3 resistance gene is not completely dominant inheritance. Tomato yellow leaf curl disease resistant breeding available PCR method for rapid screening resistance sources to aggregate multiple resistance genes in a variety, to foster a higher and more stable resistance of tomato yellow leaf curl disease tomato varieties.
