Abstract


Objective: Epilepsy is a neurological common chronic diseases, abnormal discharge of brain neurons caused by abnormal brain function characterized by clinical findings that about 1/3 of patients with epilepsy have varying degrees of cognitive dysfunction, including impaired memory , comprehension decline and decreased attention. In recent years come to realize that epilepsy patients with cognitive disorders seriously affect the patient's quality of life, so the mechanisms and treatment of cognitive impairment in patients with epilepsy is also of concern. By intraperitoneal injection of pentylenetetrazol (the pentylenetetrazol PTZ) kindling Sprague-Dawley (SD) rats, and given memantine hydrochloride epileptic rats intervention to observe the of epileptic rats memantine intervention rats learning and memory change, and the use of Western Blot, immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR) method to observe the hippocampus kinase glycogen synthase-3beta (glycogen synthase kinase 3, GSK-3beta) phosphorylation of sugar the original kinase synthase-3β (P-GSK-3β) protein expression changes in and GSK-3βmRNA, expression, to explore the role of GSK-3beta cognitive dysfunction in epileptic rats. Observed memantine hydrochloride to improve cognitive function in epileptic rats, to further explore its possible mechanism. : Created by daily intraperitoneal injection of PTZ kindling model of epilepsy. After the Y-maze screened qualified 48 SD rats (7-8 weeks, starting weight 200 ± 20 g) were randomly divided into normal control group (NC group), epilepsy alone group (PTZ group) and memantine hydrochloride intervention group (MN group, 10mg/kg). Control group of 12, with a the epilepsy group of memantine group of 18 from the experiment the first day of PTZ group and MN 9:00 am intraperitoneal injection of PTZ 35 mg / kg, the NC group at the same time intraperitoneal injection of the appropriate dose saline after each injection was observed for 1h. Morris water maze test results the navigation test results: Day 1, with NC rats escape latency comparison, PTZ group of rats escape latency was significantly longer (P lt; 0.05) MN rats escape latency was significantly longer ( P lt; 0.05), the PTZ group MN rats escape latency was no significant difference (P gt; 0.05); two days, three groups of rats escape latency compared with each other results the same as the first day; 3 days PTZ rats escape latency compared with NC group was significantly longer (P lt; 0.05), MN rats escape the incubation period and PTZ rats was significantly shorter (P lt; 0.05), the NC group and MN rats escape incubation period no significant difference (P gt; 0.05); 4 days, three groups of rats escape latency compared with each other the same as the first three days. 3 the rat hippocampus Western blot results PTZ rats P-GSK-3β protein expression levels (0.246 ± 0.115) compared with NC group (0.718 ± 0.076) and MN group (0.670 ± 0.079) was significantly lower (P lt; 0.05), NC between the group and MN group was not statistically significant (P gt; 0.05). NC group (1.210 ± 0.139), PTZ group (1.013 ± 0.046), MN group (1.164 ± 0.155) rats between GSK-3beta protein expression was no significant difference (P gt; 0.05). 4 GSK-3βmRNA NC group (1.098 ± 0.054) expression in the hippocampus of rats with PTZ group (1.149 ± 0.046) and MN (1.105 ± 0.048) compared with each other no significant difference (P gt; 0.05). 5 immunohistochemical results GSK-3β expression in rat hippocampal CA3: GSK-3beta-positive cells in the hippocampal CA3 tan, mainly expressed in the cytoplasm of cells in the hippocampus, the NC group (32.428 ± 5.223), PTZ group (31.571 ± 4.391) and MN group (32.000 ± 5.537) between the number of positive cells compared with each other no difference was not statistically significant (P gt; 0.05) Conclusion: 1 pentetrazole ignite chronic epileptic rats learning and memory impairment, epilepsy rat hippocampus in P-GSK-3β protein expression decreased the expression of GSK-3beta protein and GSK-3βmRNA, no significant change, suggesting that P-GSK- 3beta may be involved in the pathogenesis of cognitive dysfunction in epilepsy. 2 memantine hydrochloride can significantly improve the epileptic rats cognitive function, can improve the P-GSK-3β protein levels in the hippocampus, no effect on the expression of GSK-3beta protein and GSK-3βmRNA, prompted memantine hydrochloride by regulating GSK -3beta activity to improve cognitive impairment in epilepsy.
