Abstract


Pregnancy-induced hypertension, gestational hypertension (PIH), is a common syndrome in pregnancy. The disease is a serious threat to the safety of mother and child, has become the leading cause of death of the maternal and perinatal. According to the severity of pregnancy-induced hypertension, clinical will be divided into: gestational hypertension, mild pre-eclampsia, severe pre-eclampsia and eclampsia. Seriously ill pregnant women in pregnancy hypertension by taking magnesium sulfate antihypertensive drugs to control the disease. At present, the pathogenesis of pregnancy-induced hypertension is not yet clear, but the most popular explanation is that vascular endothelial injury doctrines. The doctrine, lipid peroxidation is characteristic of metabolic reactions. Based on NMR metabolomics (NMR-based Metabonomics) method, combined with nuclear magnetic resonance spectroscopy (NMR) and principal component analysis (PCA) method, exploration and analysis of severe pre-eclampsia in pregnancy hypertension disease. Pregnant women specific metabolic reactions observed pregnancy hypertension to find characteristics distinguish between pregnancy-induced hypertension pregnant women and normal pregnant women metabolites provide experimental support for the study of pathogenesis of pregnancy-induced hypertension. We collected normal pregnant women, maternal serum pregnancy-induced hypertension in pregnant women, and treatment of pregnancy-induced hypertension (maternal and umbilical blood) and placenta pregnancy hypertension metabolomics analysis. The experimental design is as follows: First, observed by NMR techniques metabolites characteristics of the three groups of pregnant women. Wherein the serum sample to be detected by conventional liquid NMR techniques; placental samples detected by the HR-MAS NMR. Secondly, multivariate statistical analysis of NMR experimental data by PCA automatic NMR pattern recognition; Finally, the results of NMR experiments and principal component analysis data integration, to draw conclusions and to give a reasonable explanation. The study found that, compared to normal pregnant women and treatment of pregnant women after pregnancy hypertension, pregnancy-induced hypertension pregnant women in the serum and placenta showed specific metabolic characteristics: significant reduction in maternal blood lipid content, choline content increased significantly; glycerol, glycerophosphate, choline and choline phosphate content also has a certain degree of rise in the placenta. A small number of placental samples of pregnant women considering pregnancy hypertension, compared with the normal group and treatment group of pregnancy-induced hypertension, metabolic differences is not obvious, we maternal plasma lipid, choline (free and lipid) , glycerol metabolites as the characteristics of pregnancy-induced hypertension in pregnant women. The amount of the samples and research data is still quite limited, in order to improve the accuracy and reliability of the results of this study, still need to improve the follow-up study based on a large number of samples.
